Self-made Gengnian Decoction For The Treatment Of Menopausal Syndrome And Study On The Mechanism Of Delaying Uterine Aging

Objective:By testing the level of E2?FSH?LH of patients who taked Gengnian decoction,we set up perimenopause model rat with deficiency of kidney-yang.Then observed the exfoliated cell of vagina?the uterus index?the pathological changes of endometrium?VEGF?ER?HSP70 and the expression of mRNA of those rats to explore the influence mechanism of Gengnian decoction on the patients of MPS.Methods:Clinical research:We selected 70 MPS patients with deficiency of kidney-yang or with deficiency of kidney-yang and kidney-yin,then we devided them randomly into treatment group and control group.The treatment group take Gengnian decoction and the control group take the textbook decoction.And to explore the difference of the two group on the level of E2?FSH?LH before treating,and the difference between groups after treating.Experimental research:We chose natural aging perimenopausal female SD rats by using vaginal exfoliate cell to determine the estrous cycle and cell measurement,and injected hydrocortisone to build the rats model with deficiency of kidney-yang.Then they were divided randomly into four groups:the model group?the textbook decoction group?Gengnian decoction group?the tibolone group;and we chose young rats of 4 months and aged rats of 18 months as the control group.All the rats of uncontrol groups were gavaged of 15 days,then they were drugged after 24 hours when they were gavaged at the last time,after that we taked their vaginal exfoliate cell and observed the MI under the micro scope.After killed rats,we removed their uterus to calculate the uterus index,observed the morphological change of uterine tissue by using HE staining,and tested the positive expression of uterine HSP70VEGF?ER?HSP70mRNA?VEGFmRNA?ERmRNA by Immunohistochemistry and in Situ Hybridization technique.Results:Clinical research:The textbook decoction and Gengnian decoction could reduce the level of FSH and LH,and increase the level of E2(P<0.05)when compared with groups before treating.And those effects of Gengnian decoction were more remarkable when compared with textbook decoction after treating.Experimental research:?The Gengnian decoction group could improve the percentage of superficial cells,and reduce the percentage of basal cells when compared with the aged group and the model group,and the difference was significant(P<0.05);but the difference between the Gengnian decoction group and the tibolone group and the textbook decoction group has no significant(P>0.05)?The index of uterus in Gengnian decoction group?the tibolone group?the textbook decoction group?the model group were increased significantly(P<0.05),and the difference between Gengnian decoction group and the tibolone group?the textbook decoction group was significant(P<0.01).?The Gengnian decoction group could improve uterine atrophy when compared with the model group(P<0.01),the effect between Gengnian decoction group and the tibolone group has no difference(P>0.05),but make a big difference when compared with the textbook decoction group(P<0.05)?The Gengnian decoction group?the tibolone group?the textbook decoction group could up-regulate the Positive Expression ofER?ERmRNA?VEGF?VEGFmRNA(P<0.05),but the level of the textbook decoction group was lower when compared with the Gengnian decoction group and the tibolone group(P<0.05).There has no significant differece between the tibolone group and the Gengnian decoction(P>0.05).?The Gengnian decoction group and the tibolone group could up-regulate the Positive Expression of HSP70 and HSP70mRNA when compared with the textbook decoction group and the model group(P<0.05),but the effect between he Gengnian decoction group and the tibolone group was similar(P>0.05).Conclusion:Clinical research:The Gengnian decoction and the textbook decoction had benign regulative effect on FSH?LH?E2 of MPS patients with deficiency of kidney-yang or with deficiency of kidney-yang and kidney-yin,and the effect of the Gengnian decoction was more remarkable.Experimental research:The Gengnian decoction can improve the MI of the aginal epithelial cells and the index of uterus,and have the estrogen-like effects.And maybe through this effect can up-regulate the expression of ER?VEGF?HSP70?ERmRNA?VEGFmRNA?HSP70mRNAon the uterine,and postpone the degenerative change of structure and function of the uterine.And supply abundant blood to ovary to maintain the normal physiological function,so that to treat MPS.
