As an important part of Chinese medicine theory system,the theory of'five-zang-viscera corresponding to four seasons' appeared in Inner Canon of Yellow Emperor firstly.The theory contains mainly 'the liver concected with spring','the heart concected with summer','the spleen concected with long-summer','the lung concected with autumn','the kidney concected with winter' and it's an important part of Chinese medicine theory system,it mainly reflects the inherent thinking mode of the TCM theory "Correspondence between Man and Nature".This theory holds that the human five internal organs Physiological and pathological changes in a certain extent,affected by the nature of climate change.As an important part of the theory of'five-zang-viscera corresponding to four seasons',the theory of 'the liver concected with spring' holds that the liver belongs to wood,is lush in spring and concected with spring,which the physiological function of organism in spring concects with the nature of climate change in spring closely and it mainly reflects the aspect to the influences of'Liver controlling dispersion'.This issue sthdies the theory of'the liver concected spring' through a series of experiment and deepens the understanding to heory of 'five-zang-viscera corresponding to four seasons' in TCM further.1 ObjectiveThe topic wants to study the influences of the seasonal changes to the gene expression in mRNA of SRB ? and BSEP in liver from the perspective of physiology and pathology,so as to provide a new idea and same experimental basis for the related research of 'five-zang-viscera corresponding to four seasons' theory and the function of Liver Controlling Dispersion.2 Methods2.1 Exprimental animalsOne hundred and twenty health male SD rats,clean,weighted 170-190g,brought seven days before beginning of vernal equinox,summer solstice,autumnal equinox,winter solstice in 2015,adaptive feed seven days,weighing,were randomly divided into three groups:normal group,CUMS group,bondage group,there were ten rats in every group.Normal group received natural light,ate and drank freely,and were controlled under certain room temperature,CUMS group and bondage group are operated 21 days.All the rats would be weighted before being beheaded.2.2 Test MethodsThe gene expression in mRNA of SRB I and BSEP in liver used Real-Time PCR of TaqMan method to test.2.3 Analysis methodThe differences of groups were tested by SPSS 17.0.The values were calculated with One-Way ANOVA test,p<0.05 meant obvious difference,p<0.01 meant significant differences.All the data were showed by mean ? standard.3 Results3.1 Influences on the gene expression in mRNA of SRB I and BSEP in liver by Liver controlling dispersion(1)Spring:The gene expression of SRB I in mRNA in liver trends to normal group>CUMS group>CRS group,the BSEP gene trends to CRS group>CUMS group>normal group,and there was a significant difference(p<0.05)in normal group compared with CRS group of BSEP gene.(2)Summer:The gene expression of SRB I and BSEP in mRNA in liver all trends to CUMS group>CRS group>normal group,and there was a very significant difference(P<0.01)in normal group compared with CRS and CUMS group.(3)Autumn:The gene expression of SRB I in mRNA in liver trends to CRS group>normal group>CUMS group,the BSEP gene trends to CUMS group>CRS group>normal group,and there was no significant difference(P>0.05)in normal group compared with the other groups.(4)Winter:The gene expression of SRB I and BSEP in mRNA in liver all trends to CUMS group>normal group>CRS group,and there was a significant difference(p<0.05)in normal group compared with CUMS group of BSEP gene.3.2 Influences on the gene expression in mRNA of SRB I in liver by seasonal variation(1)Physiological group:The gene expression in mRNA of SRB ? in liver of Physiological group hand trend winter>spring>autumn>summer,and the winter are higher than the others obviously.Besides,there was a very significant difference(p<0.01)in spring,summer,autumn compared with winter.(2)CUMS group:The gene expression in mRNA of SRB ? in liver of CUMS group hand trend winter>spring>summer>autumn,and the winter are higher than the others obviously.Besides,there is a very significant difference(p<0.01)in spring,summer,autumn compared with winter.(3)CRS group:The gene expression in mRNA of SRB ? in liver of CRS group hand trend winter>spring>autumn>summer,and the winter is higher than the others.Besides,there was a very significant difference(p<0.01)in autumn,summer compared with winter;there also was a significant difference(p<0.05)in spring compared with winter.3.3 Influences on the gene expression in mRNA of BSEP in liver by seasonal variation(1)Physiological group:The gene expression in mRNA of BSEP in liver of Physiological group hand trend winter>spring>autumn>summer,and the winter is higher than others obviously.Besides,there was a very significant difference(P<0.01)in spring,summer,autumn compared with spring and winter;there also was a significant difference(p<0.05)in spring compared with summer.(2)CUMS group:The gene expression in mRNA of BSEP in liver of CUMS group hand trend winter>spring>sutumn>summer,and winter is the highest,the others are no big difference.Besides,there was a very significant difference(p<0.01)in winter compared with the other three seasons.(3)CRS group:The gene expression in mRNA of BSEP in liver of CRS group hand trend spring>winter>autumn>summer,and spring is the highest,summer is the lowest,it is different from the other groups.Besides,there was a very significant difference(P<0.01)in summer compared with spring;there also was a significant difference(p<0.05)in spring compared with autumn,summer compared with winter.4 Conclusions(1)In normal and Physiological group,the gene expression in mRNA of SRB I and BSEP in liver will change by seasons and most of them have significant difference.So it reflect that the function of Liver controlling dispersion can change with the seasonal change on one hand.On the other hand,it also reflect that the seasonal changes can influence the gene expression in mRNA of SRB I and BSEP in liver.(2)In the same season,the CUMS and CRS method can make effect on the gene expression in mRNA of SRB I and BSEP in liver,and this influence behaves obviously in summer and autumn.(3)Although the CUMS and CRS method are both the animal model of depression,there are difference for the gene expression in mRNA of SRB I and BSEP in liver of animal model,so it can also display out that the CUMS and CRS model are not the same completely.
