Objective: To observe the effect of Chinese acupoints selection treatment of fixed local selection of acupoint for compatibility of different acupoints,and to explore the effect of selection of acupoints by acupuncture on compatibility of acupoints.Methods: 60 SD rats were randomly divided into normal control,model,“Bahui”(GV 20)-“Shenmen”(HT 07,3-acupoints)group,“Bahui”(GV 20)-“Sanyinjiao”(SP 06,3-acupoints)group,“Bahui”(GV 20)-non-acupoint(3-acupoints)group,with 12 rats in each group.With continuous abdominal cavity injection of p-chlorophenylalanine(PCPA,500 mg / kg,100 mg / ml)2d the insomnia model was established.“Bahui”-“Shenmen” group,“Bahui”-“Sanyinjiao”group,“Bahui”-non-acupoint group were with one time daily acupuncture,each for 30 minutes with continuous 7 days treatment.The levels of MT in the SCN,VLPO and plasma and were detected by ELISA,and the expression of Clock mRNA and Bmal1 mRNA in the ventrolateral preoptic area(VLPO)and suprachiasmatic nucleus(SCN)of the hypothalamus were detected by real-time fluorescence quantitative PCR(Real time-PCR).Results:1?After modeling,pentobarbital righting the experimental results show that the model group the sleep latency than before modeling extension(P<0.05),sleep duration was shortened before modeling(P<0.05),and to compare the difference with the control group(P<0.05),suggesting that the model was successful.Compared with normal group,the sleep latency period of sodium pentobarbital sleep was increased in the model group(P<0.05),and that of sleep duration.Compared with model group,the sleep latency period was decreased in the same meridian group,related meridian group,non-acupoint group(P<0.05),and the sleep duration was increased in the same meridian group(P<0.05).2?Diet: compared with the blank group,the model group to reduce consumption,the difference was statistically significant(P<0.05);compared with the model group,Baihui plus Shenmen group increase food intake(P<0.05);comparison between treatment groups,Baihui plus Shenmen group than Baihui and Sanyinjiao group,Baihui and non acupoint group(P<0.05).The amount of drinking water: model group was less than that of control group(P<0.05);Baihui plus Shenmen group,Baihui and Sanyinjiao group,Baihui and non acupoint group less than the model group(P<0.05);the treatment group were no difference(P>0.05).3?The body weight increase of the rats compared with the control group,model group,body weight growth quantity is very few,the difference was statistically significant(P<0.05);compared with the model group,Baihui plus Shenmen group,Baihui and Sanyinjiao group weight increase more(P<0.05);comparison between treatment groups,Baihui plus Shenmen group than Baihui and Sanyinjiao Baihui +group,non acupoint group(P<0.05).4?The levels of MT of the ventrolateral preoptic area of hypothalamus nucleus were higher in the same meridian group than the model group and non-acupoint group(P<0.05)while lower than blank control group(P<0.05).In the suprachiasmatic nucleus,levels of MT were higher in the same meridian group than model group and the non-acupoint group(P<0.05).The levels of MT in plasma were higher in the same meridian group than the model group,related meridian group and non-acupoint group(P<0.05)while lower than blank control group(P<0.05).5?Comparing the expression quantity of Clock mRNA and Bmal1 mRNA in the SCN and VLPO regions: compared with the blank group,the expression quantity of model group was all reduced(P <0.01).Compared with the model group,expression quantity of Clock mRNA was increased in “Bahui”-“Shenmen” group,“Bahui”-“Sanyinjiao” group(P <0.01);the expression quantity of Bmal1 mRNA in“Bahui”-“Shenmen” group,“Bahui”-“Sanyinjiao” group,“Baihui”-non-acupoint group was improved(P <0.05).Compared among the treatment groups,the expression quantity of “Bahui”-“Shenmen” group was higher than that of “Bahui”-“Sanyinjiao” group and “Bahui”-non-acupoint group(P<0.01);the expression quantity of Clock mRNA in “Bahui”-non-acupoint group was lower than that in“Bahui”-“Sanyinjiao” group(P <0.01).Conclusion:1?The acupuncture treatment of the fixed brain local “Baihui” acupoint with compatibility of “Shenmen” acupoint can improve the sleep time and latency of insomnia rats.2?Fixed brain local “Baihui” acupoint with compatibility of “Shenmen”acupoint can improve the expression level of biological Clock mRNA and Bmal1 mRNA,and enhance the content of melation.3?Verified that the selection of acupoints by acupuncture can improve compatibility of acupoints effect.
