Experimental Study On The Effect Of Weifu Recipe On The Expression Of BGC-823 Xenografts And PPAR? Gene Expression In Nude Mice

OBJECTIVE: observing the effect of Weifufang on gastric cancer BGC-823 xenografts in nude mice and the activity of peroxisome proliferator-activated receptor ?(PPAR?)gene.To explore the possible mechanism of Weifufang in inhibiting BGC-823 transplanted tumor in nude mice.Methods: The mices of gastric cancer BGC-823 xenografts were treated with different concentrations of Weifufang.The tumor size of the transplanted tumor was observed with different doses of Weifufang.The histopathological changes of the tumor were observed by HE staining,The expression of PPAR? gene was detected by immunohistochemistry and Western blot Methods.RESULTS: Weifufang could inhibit the growth of BGC-823 xenografts in nude mice by two weeks.The inhibition rate of 5-fu positive control group,low-dose Weifufang group,middle-dose Weifufang group and high-dose Weifufang group were: 30.67%? 19%? 49.52%? 29.36%.The IOD value of PPAR-?was detected by immunohistochemistry in Distilled water group,the5-fu positive control group,low-dose Weifufang group,middle-dose Weifufang group and high-dose Weifufang group were 0.04± 0.02 ? 0.05±0.01?0.05± 0.01?0.09± 0.02?0.06± 0.01.There was significant difference between each group with middle-dose Weifufang group(p <0.05).The expression of PPAR-?was detected by Western blotting in Distilled water group,the 5-fu positive control group,low-dose Weifufang group,middle-dose Weifufang group and high-dose Weifufang group were0.3160±0.0136 ? 0.7240±0.0181 ? 0.5640±0.0199 ? 0.7180±0.0289 ?0.5700±0.0200.There was significant difference between distilled water group and each group(p <0.05).There was no significant difference between the 5-fu group and the middle-dose Weifufang group(p> 0.05).There were significant difference between 5-fu group and the low-dose Weifufang group and high-dose Weifufang group.There was significant difference between the middle dose Weifufang group and low and high dose Weifufang group(p <0.05).Conclusion: 1?Weifufang can inhibit the growth of gastric cancer BGC-823 xenograft in nude mice;2? Weifufang could inhibit the growth of gastric cancer BGC-823 tumor by up-regulating the expression of PPAR?gene,and the effect was best in middle dose group.
