| objective:By electro acupuncture Depression model rats with gastric motility disorder at Zusanli,Taichong,Ganshu,Weishu etc,observe its behavior,gastric emptying,brain tissue DA,the changes of SP and CGRP of gastric motility and brain hippocampus DA in serum,to explore the part of mechanism of Electroacupuncture in depression model rats with depressive symptoms and the gastric motility disorder intervention.Method:60 SD rats,Randomly selected 16 as the blank group,The other 44 SD rats were prepared by the method of chronic unpredictable mild stress preparation of depression model,CUMS consists of 7 different stimulus:Fasting for 24 hours;the water deprivation for 24 hours;the light and shade upside down for 24 hours;the wet cages for 24 hours;the tail clamp for 3 minutes;30 minutes,the violent shaking;forced swimming for 5 minutes.1 stimulu randomly selected each day,the past 2 days do not use the same stimulus,modeling time of 4 weeks.To determine the success of the rat model of depression by behavioral observation;At the end of the molding,60 SD rats,including the blank group,were used to determine whether the depression model rats had gastric motility disorder,After removing 12 rats that did not meet the requirements from the model group,The blank group and the model group were randomly selected 8.Gastric emptying was measured in 16 rats,to further verify whether the rats with gastric motility disorder,The remaining 24 rats from the model group were randomly divided into three groups:Acupuncture group and fluoxetine group,model group,8 rats in each group.Blank group:conventional method of feeding,do not do any treatment;Model group:conventional method of feeding,only do binding treatment;Acupuncture group:conventional method of feeding,Acupuncture at "Zusanli","Taichong","Ganshu ","Wei Shu","Zhongwan"point,The "Zusanli","Taichong","Gan Shu","Wei Shu" points by electro acupuncture,Bilateral Zusanli,Taichong as a group,Gan shu,wei shu as a group,Zhongwan only acupuncture,no electricity;Prozac group:conventional method of feeding,Prozac intragastrically.Adult dose of 20 mg/day,The animal dose is about 0.018(200 g rats and human body surface area ratio of 70 kg),The concentration of 0.36mg/mL prepared Prozac fluoxetine hydrochloride suspension,Shake well before use,The volume of intragastric administration was lmL/200g body mass,once a day,continuous administration for 14 days.After intervention,4 groups of animals including the blank group were killed for Gastric emptying test;Abdominal aorta blood collection,Detection of SP and CGRP in serum;Rapid extraction of brain,Rinse with normal saline,After removal of surface water,100%formalin fixed,Detection of dopamine(DA)in hippocampus.Results:1.The effect of Electro acupuncture on behavior of depression model ratsThe changes of horizontal displacement of open field experiment in rats:There was no significant difference in the total displacement of the experimental rats in the open field experiment before the model was established,After modeling,the horizontal motion of the total displacement of the model group,fluoxetine group and acupuncture group was significantly lower than that of the control group,the difference was statistically significant(P<0.01);After the intervention,Compared with the horizontal displacement of the blank group,the model group was significantly lower,the difference was statistically significant(P<0.01),the acupuncture group was significantly lower.the difference was statistically significant(P<0.05).Prozac group had no statistical significance(P>0.05);Compared with the model group,acupuncture group and fluoxetine group the total displacement of the horizontal direction increased significantly,the difference was statistically significant(P<0.01);Compared with fluoxetine group,acupuncture group total horizontal displacement decreased slightly,the difference was not statistically significant(P>0.05).The changes of the number of vertical movement in open field experiment in rats:There was no significant difference in the number of vertical movement in the open field experiment in each group before the model was established,After modeling,the vertical movement of the model group,fluoxetine group and acupuncture group significantly lower than that of the control group,the difference was statistically significant(P<0.01);After the intervention,the model group,fluoxetine group and acupuncture group the number of vertical movement is significantly lower than the control group,the difference was statistically significant(P<0.01);Compared with the model group,acupuncture group and fluoxetine group significantly increased the number of vertical movement,the difference was statistically significant(P<0.01);compared with fluoxetine group,acupuncture group,the number of vertical motion is slightly lower,the difference was not statistically significant(P>0.05).2.Effect of Electroacupuncture on gastric emptying in ratsThe results of ECG detection:Compared with the blank group,the slow wave amplitude of rats in the model group was significantly lower,the difference was statistically significant(P<0.05);The percentage of normal slow wave is significantly lower,the difference was statistically significant(P<0.01),no gastric electric rhythm percentage is significantly higher,the difference was statistically significant(P<0.01);bradygastria percentage is significantly higher,the difference was statistically significant(P<0.01),tachygastrias percentage is high,the difference no statistical significance(P>0.05).Gastric emptying rate:The model group compared with blank group,the gastric emptying rate significantly lower,the difference was statistically significant(P<0.05);compared with the model group,acupuncture group,the gastric emptying rate was significantly higher,the difference was statistically significant(P<0.05),gastric emptying rate of Prozac group,there was no statistically significant difference(P>0.05);compared with Prozac group,acupuncture group,the gastric emptying rate increased significantly,the difference was statistically significant(P<0.05).3.Effect of Electroacupuncture on DA in hippocampus of ratsThe average optical density of DA in hippocampus of rats:The average optical density of DA in model group was significantly lower than the blank group,the difference was statistically significant(P<0.05);compared with the model group,acupuncture group and fluoxetine group in the hippocampus DA average optical density value is significantly higher,the difference was statistically significant(P<0.05);acupuncture group compared with fluoxetine group,hippocampus DA the average optical density had no significant difference.The results of brain tissue slices in brain tissue:The blank group(Figure 1)cell morphology integrity,cell membrane and nucleus clear boundaries;the model group(Figure 2)the visible portion of irregular cells,cytoplasm and nucleus is not clear,color than the normal group cells is deep;Prozac group(Figure 3)shows a small part of irregular cells,significantly reducing the number of a model group,and the degree of staining the cells in the control group had no significant difference;the acupuncture group(Figure 4)shows small irregular cell number less than the model group,the degree of color and the cells in the control group had no significant difference.4.Effect of Electroacupuncture on SP and CGRP in serum of model ratsDetection of SP in serum by Elisa:Compared with the blank group,model group,fluoxetine group and acupuncture group SP content decreased significantly,the difference was statistically significant(P<0.01);compared with the model group,acupuncture group,SP content increased significantly,the difference was statistically significant(P<0.01),Prozac group SP was significantly reduced.the difference was statistically significant(P<0.01);compared with fluoxetine group,acupuncture group,SP content increased significantly,the difference was statistically significant(P<0.01).Detection of CGRP in serum by Elisa:Compared with the blank group,model group,fluoxetine group and acupuncture group CGRP content increased significantly,the difference was statistically significant(P<0.01);compared with the model group,acupuncture group,CGRP content decreased significantly,the difference was statistically significant(P<0.01),Prozac group CGRP was significantly increased,the difference was statistically significant(P<0.01);compared with fluoxetine group,acupuncture group,CGRP content decreased significantly,the difference was statistically significant(P<0.01).Conclusion:1.Acupuncture can obviously improve the depressive symptoms and the gastric motility disorder in the depression model rats,It may be achieved by increasing the content of DA in the hippocampus and serum SP and reducing the content of CGRP.2.Acupuncture can improve the depression symptoms of rats with depression and improve the symptoms of gastric motility disorder.Prozac can improve the depression symptoms of rats with depression may aggravate the gastric motility disorder.Therefore.Acupuncture in improving depression rat model of depressive symptoms and gastric motility disorder than Prozac has obvious advantages. |
