| Objective:Based on the fact that the disorder of the Blood Glucose-Axis HPA-Circadian Rhythm mood exists in diabetic rats with depression and acupuncture could adjust the disorder of he Blood Glucose-Axis HPA-Circadian Rhythm mood.Using the method of studying the today’s timing biology, Observation study of the animals’movements and the technology of molecular biology and etc. With those method,studying the changing rhythm of the Blood Glucose-Axis HPA-Circadian Rhythm mood of diabetic rats with depression.studying the influence of the expression of genes of PerlmRNA and Per2mRNA,and uncovering the principle of molecular biology about using acupuncture to adjust the the disorder of the Blood Glucose-Axis HPA-Circadian Rhythm mood of diabetic rats with depression.Methods:Putting 32 healthy SD male rats into 4 groups equally at random.Blank group has 8 rats,diabetic group has 8 rats,diabetic with depreslsion group has 8 rat and the acupuncture group has 8 rats.Giving the normal feeding to Blank group.Giving the fatty fodder and injecting STZ reagent i.p. to the rats to the diabetic group.Besides the management of the diabetic group,restrain management is also used to the diabetic with depression group.Besides the management of the diabetic with depression group, acupuncture management is used to the acupuncture group,the detailed method is picking Tsusanli point, Sanyinjiao point and Baihui point, needling those points,and keeping this state for 15 minutes.This treatment will be continued for 7days.After the treatment.we will get the blood serum and brain tissue by taking blood from Arteria femoralis and cutting the neg of rats.Using the ELISA to assay the CORT.Using the PC R technology to check the gene expression of Per1,Per2 etc.Result:1, (1)Compared to the blank group rats,the blood glucose of the diabetic group rats have increased(P<0.05) and the result about the open field test of the diabetic group rats have decreased(P<0.05). (2) Compared to the blank group rats,the maximum and the minimun of the blood glucose in 24 hours of the diabetic group rats are different (P<0.05), and the time of the maximum blood glucose and the result about the open field appearing has been delayed.2, (1)Compared to the blank group and the diabetic group rats,the blood glucose of the diabetic with depreslsion group rats have increased(P<0.05) and the result about the open field test of the diabetic with depreslsion group rats have decreased(P<0.05). (2) Compared to the blank group rats,the maximum and the minimun blood glucose in 24 hours of the diabetic with depression group rats are different (P<0.05), and the time of the maximum blood glucose appearing has been delayed,and the changing tendency of the result about the open field test of the diabetic with depression group rats in 24 hours is nearly contrary to the blank group rats.3, Compared to the blank group and the diabetic with depression group rats,the blood glucose of the diabetic with depression group rats have decreased(P<0.05) and the result about the open field test of the acupuncture group rats have increased(P<0.05). (2) Compared to the blank group rats,the maximum and the minimun of the blood glucose in 24 hours of the acupuncture group rats are different (P<0.05),but the time of the secondly maximum and secondly minimum blood glucose appearing are same to the time of the maximum and the minimum blood glucose appearing of the blank group, and the blood glucose are nearly same to the blank group without the big oscillations from 10:00-22:00 in 24 hours(P>0.05).4, Compared to the blank group rats,the other three group rats have the higher level of the CORT(P<0.05),and the level of the other three group compared:the diabetic with depression group> the diabetic group>the acupuncture group(P<0.05).5, Compared to the blank group rats,the diabetic group rats and the diabetic with depression group rats have the higher gene expression of Per2mRNA(P<0.05),and compared to the he diabetic group rats and the diabetic with depression group rats,the acupuncture group rats have the lower gene expression of Per2mRNA(P<0.05) Compared to each other,the gene expression of PerlmRNA of the rats,the blank group< the acupuncture group<the diabetic group<he diabetic with depression group.Conclusion:1, Acupuncture has the function of the ameliorating the blood glucose and depression of the diabetic rats with depression, and it also has theunction of the ameliorating the allorhythmia of blood glucose and the emotion.2, Acupuncture has the function of decreasing the level of the CORT.3, Acupuncture has the function of decreasing the gene expression of Per2 mRNA and it may by the means of adjusting the gene expression of Per2 mRNA, then ameliorating the allorhythmia of blood sugar level and the emotion, thus cure the diabetic with depression. |
