| Objective:The research aims to discuss the effects of electro-acupuncture and catgut-implantation regulate circadian rhythm of morphine-addiction mice and gene expression of Perl and Per2in suprachiasmatic nucleus (SCN) in order to provide experimental basis to the clinical application of electro-acupuncture and catgut-implantation therapy of morphine addiction.Methods:42male C57BL/6J mice were domesticated and selected, and then randomly divided into4groups which were control group(n=14), model group(n=14), electro-acupuncture(n=7) and thread embedding group (n=7). Inject morphine hydrochloride injection incrementally under skin in seven days what were20mg/kg,40mg/kg,60mg/kg,80mg/kg,100mg/kg,100mg/kg,100mg/kg to build models, when the control group was injected normal saline in the same proportion (8am,8pm). Meanwhile, fifteen minutes after injection, the electro-acupuncture group was under treatment by administering electro-acupuncture at bilateral Shenshu point(BL23) with HANS Acupoint and Nerves Stimulator(HANS-2000) using2/15Hz and0.2mA for15min twice a day, and the catgut-implantation group was implanted chromic catgut of0.5centimeter at bilateral Shenshu acupoint at8am in the first day of modeling. Six days later, all the groups were observed the abstinence symptoms for one hour and given a mark after nabxone injection through abdominal cavity at10pm. At last, samples of SCN were removed and measured by RT-PCR method to test the gene expression level ofperiod genes Perl and Per2.Results:1. Behavioral results â‘ Withdrawal symptom:After naloxone administration, the control group had no significant withdrawal symptoms. The model group was excited during the first hour after naloxone, and developed withdrawal symptoms such as jumping, the wet dog sample trembling, diarrhea, et al, all the weight lost were more than two percent. The final scores of model group had significant difference with the control group(p<0.05). The electro-acupuncture group and catgut-implantation group also had withdrawal symptoms, and it had significant difference with the model group(p<0.05).â‘¡Weight in seven days:There were no significant differences in every group before the experiment(p>0.05). When building models started, the control group was normal as usual The model group had dramatic decline in weight during seven days, and the differences had statistical significance compared to control group(p<0.05). There were downtrends in weight of electro-acupuncture group and catgut-implantation group what had statistical significance compared to control group(p<0.05), and in contrast with the model group, from the sixth and seventh day separately, there were upward trend but had no significant(p>0.05).â‘¢The circadian rhythm characteristics:There were no significant differences in period, amplitude, and activity among three groups before the experiment(p>0.05). After the study began, the control group mice were normal as before, when the model group had obvious change in amplitude and activity compared to the control group(p <0.05), except period(p>0.05). The electro-acupuncture group mice had distinct changes in amplitude and activity compared with control group and model group(p<0.05), except period(p>0.05). The catgut-implantation group mice had significant changes in amplitude (p<0.05), except period and activity (p>0.05) compared to control group and model group.2. The Per genes expression:The data of RT-PCR indicated that the gene expression levels of Perl and Per2in SCN of morphine-addiction mice were down-regulated compared to the control group and it had statistical significance(p<0.05). Compared with the control group, the electro-acupuncture group mice had significant difference in gene expression levels of Perl and Per2(p<0.05), when Per2gene expression were up-regulated in contrast with model group(p<0.05), but Perl gene had no significant difference(p>0.05). Compared with the control group, the catgut-implantation group mice had significant difference in gene expression levels of Perl and Per2(p<0.05), when Perl gene expression were up-regulated in contrast with model group(p<0.05), meanwhile Per2gene had no significant difference(p>0.05).Conclusion:(1)Both of electro-acupuncture and thread embedding treatment could relieve withdrawal symptoms, reduce the score of withdrawal symptoms and weight downtrend of morphine addiction mice.(2)Both of electro-acupuncture and thread embedding treatment could improve the circadian rhythm disorder. The electro-acupuncture group mainly manifests in reducing amplitude and activities, and the thread embedding group mainly shows in reducing amplitude.(3)Addictive behaviors were related to genes expression of Perl and Per2. In order to relieve withdrawal symptoms, we could up-regulate the genes expression of Perl and Per2. |
