Separation, Purification And Preliminary Study Of The Activity Of Lycopene

Prodigiosins,a secondary metabolite biosynthesized by some Actinomycetes,bacteria,possesses a common pyr-rolylpyrromethene(PMM)skeleton.Prodigiosin is of great interest due to its antibacterial,antifungal,antimalarial properties,immunosuppressive activity,etc.Recently,lots of researches reveal that prodigiosin has great potential for applications in cancer treatment,so the research of prodigiosin attracts increasing attention.The experi,nent take strains HUB-I-107 as original strain,optimize biological synthesis of prodigiosin pathway precursors(amino acids)and purify prodigiosin,at the same time,investigate activities of prodigiosin.According to prodigiosin biosynthesis pathway using response surface methodology to optimize a number of possible precursors of HUB-I-107 prodigiosin biosynthesis pathway.By Plackett-Burman design and response surface analysis,the best concentration of factors were determined:glycine(gly),aspartate(asp),proline(pro)concentrations were 1.534629 g/L,1.469015 g/L,6.284922 g/L,the prodigiosin production was increased to from 295 mg/L to 878 mg/L,production increased three-fold.According to prodigiosin presence proportion in fennentation broth,use macroporous resin adsorption and organic solvent extraction method combination,silica gel column and C18 column were used to purify,the use of TLC to track test compounds,prodigiosin were isolated,and use like UV,MS,HNMR structural identification.Last,by the Oxford cup method,MTT method prodigiosin study inhibition bacterial and tumor cell proliferation,experimental results show that:Gram-positive and negative bacteria are inhibited by prodigiosin,the inhibition of Bacillus subtilis is the strongest effect;and prodigiosin on the MDA-MB-231 cells(breast cancer)has tumor cell proliferation inhibition,IC50 value of 0.72 ?g/mL.Prodigiosin on the tumor cells was time-dose dependent manner.
