Purpose : To demonstrate the mechanism of Jianpi Decoction intervening on atherosclercsis Apo E-/- mice in early stage by detecting from myocardial of5-hydroxytryptamine signaling pathway in based on the “Inter-relation between heart and spleen” theory, and provide experimental evidence for intervening on atherosclerosis in clinical research.Material and method: Thirty 9-week-old SPF male Apo E-/- mice adaptive feeding for one week, were randomly divided into model group, western medicine group, Jianpi Decoction group, n=10, fed with high-fat-feedstuff, while set up the normal group with the same genetic background ten male C57BL/6N mice normal feeding. Normal group and model group were gavaged with normal, the western medicine group with doses of atorvastatin calcium solution and Jianpi Decoction group with Jianpi Decoction solution for 12 weeks.Aorta and myocardial tissue were fixed and embedded in paraffin and sliced into 5um slides.Hematoxylin staining was performed to evaluate the condition of aorta and myocardium.ELISA was performed to examine the content of 5-HT and 5-HIAA in serum and myocardial tissue. Immunohistochemistry was performed to detect the protein expression of 5-HT,5-HT2 BR, PKC and NF-?B in myocardial tissue, images were shot, then used BL-2000 digital image analysis system to measure the gray value of pathological section. All the data analysised by SPSS17.0 software.Results:1. Histological morphological data shows in normal group that endothelial cells appears complete structure with normal form of internal elastic membrane and medium film elastic fiber, Smooth muscle cell(SMC) arrangement rule, no obvious pathological changes in sum.The model group: disorder SMC, apparent and thick inner membrane, broken elastic fiber,with a large number of inflammatory cell infiltration, endothelial cell lipid infiltration, and plaque formation in the intima. The western medicine group: the structure of the arterial recovered, little inflammatory cells, regular endothelial cells and SMC, smooth inner membrane, complete elastic fibers. The Jianpi Decoction group: the structure of the arterial recovered, with smooth inner membrane, complete elastic fibers and endothelial cells, regular SMC, and scattered inflammatory cells.2. Histological morphological data shows in normal group that the structure with clear intercalated discs, close cardiac muscle cells, clear cytoplasm, no inflammatory cells. The model group: disorder cardiac muscle cells, disappearing myocardial cell structures, unclear cytoplasm, with a large number of infiltrating inflammatory cells. The western medicine group: the structure of myocardial cell recovered, discrete cardiac muscle cells, accompanied with some of the clear cytoplasm, and scattered infiltrating inflammatory cells. The Jianpi Decoction group: the structure of myocardial cell recovered, closely cardiac muscle cells,clear cytoplasms, and scattered infiltrating inflammatory cells.3. ELISA date shows, by comparison with the contents of 5-HT and 5-HIAA in serum and myocardial tissue of the normal group, the ones of the model group increase(P<0.05), but5-HT conversion rate decrease(P<0.05). By comparison with the ones of the model group,the contents of 5-HT and 5-HIAA in the mice's serum and myocardial tissue of the western medicine group and Jianpi Decoction group decrease(P<0.05), 5-HT conversion rate decrease(P<0.05), the Jianpi Decoction group is superior to the western medicine group(P<0.05).4. The results of 5-HT, 5-HT2 BR, PKC, NF-?B of the gray values in the mice's myocardial tissues show that, the gray values is inversely proportional to the positive expression, that is to say, the positive staining is the deeper, the lower the gray value is. By comparison with the positive expression of the normal group, the one of the model group raises(P<0.05); by comparison with the one of the model group, the positive expression of western medicine group and Jianpi Decoction group decreases(P<0.05), the positive expression of the Jianpi Decoction group is superior to the one of the western medicine group(P<0.05).Conclusion:1. The imbalance of peripheral 5-HT in the formation and metabolism may be one Pathogenesis of atherosclerosis induced by inflammation.2. The mechanism of Jianpi Decoction on atherosclerosis may be related to the content of 5-HT and 5-HIAA of serum and myocardium, and be related to the 5-hydroxytryptamine signal pathway mediated by 5-HT2 B receptor for inflammatory reaction in the myocardium,this may be one of Jianpi Decoction ' protective effects on myocardial morphology.
