| Objective: to study the impact that fulingxingrengancao decoction combined with juzhijiang decoction Acting on the acute myocardial ischemia in rats.By observing the changes of ECG?LDH?CK-MB in rats,to discuss the curative effect of Chinese classic herbal formula in acute myocardial ischemia,and by observing apoptosis in rat,to investigate the mechanism that fulingxingrengancao decoction combined with juzhijiang decoction Acting on the acute myocardial ischemia in rats.Methods:50 SD rats, were randomly divided into normal control group, acute myocardial ischemia model group, low dose group of Chinese medicine,high dose group 0f Chinese medicine and Xikang Group with 10 rats in each group?Groups were given relevant drugs for a week,After lavage for 15 min within the last three days of the experiment,each group were given isopropyl adrenaline(every 2mg/kg) by intraperitoneal injection caused by acute myocardial ischemia model,except normal saline for Blank control group. After giving drugs by lavage for one week,collecting electrocardiogram information,testing LDH?CK-BM of zhe rat, and take out the heart,observing the myocardial cell apoptosis and Bax, Bcl- 2 protein related apoptosis expression by the DNA- end labeling method and immunohistochemical method.Results:1.ECG:one week later,test groups of electrocardiogram(ecg) in rats, electrocardiogram J piont?T wave of blank group rats did not see obvious upside down,the one of other groups can see different degrees of change,the J piont? T wave degree of acute myocardial ischemia model group is highest(P<0.05),the other two groups are betweena cute myocardial ischemia model and high dose group 0f Chinese medicine.2.LDH:one week later,In addition to the blank group, other groups have different degrees of lactic dehydrogenase,statistically significant differences in comparison with the blank group(P< 0.05),LDH of acute myocardial ischemia model group is highest,and high dose group 0f Chinese medicine is lowest(P<0.05),he other two groups are betweena cute myocardial ischemia model and high dose group 0f Chinese medicine(P<0.05).3.CK-MB:one week later,In addition to the blank group, other groups have different degrees of CK-MB,statistically significant differences in comparison with the blank group(P< 0.05),CK-MBof acute myocardial ischemia model group is highest,and high dose group 0f Chinese medicine is lowest(P<0.05),he other two groups are betweena cute myocardial ischemia model and high dose group 0f Chinese medicine(P<0.05).4.Apoptosis and apoptosis related gene :Microscopic results show that the blank group can see a small positive apoptotic cells,but did not see obvious necrosis, degeneration of myocardial cells.Model group have a large number of apoptosis of myocardial cells,apoptosis of drug group were significantly reduced in compare with the model group(P<0.05),high dose group 0f Chinese medicine reduced more obviously in compare with low dose group 0f Chinese medicine and xinkang group(P<0.05).Compared with the blank group, apoptosis and Bax ?Bcl- 2 protein expression of model group increased obviously(P<0.01),Compared with the model group,apoptosis and Bax protein expression of drug group reduced obviously(P<0.05),Bcl- 2 protein expression increased obviously(P<0.05).high dose group 0f Chinese medicine inhibiting apoptosis obviously,statistically significant differences in comparison with low dose group 0f Chinese medicine and xinkang group(P<0.05),statistical have no significance between low dose group 0f Chinese medicine and xinkang group(P>0.05).Conclusion:1.fulingxingrengancao decoction combined with juzhijiang decoction can impove the ECG of rats with acute myocardial ischemia, Reduce the content of LDH and CK-MB.2.fulingxingrengancao decoction combined with juzhijiang decoction can reduce the apoptosis rate to achieve the purpose of inhibiting myocardial apoptosis by raising the Bcl- 2 protein expression and cut Bax protein expression,to have the effect of anti myocardial ischemia. |
