Objective In this study,rheumatoid arthritis induced by collagen(Collagen-induced,arthritis,CIA)is model,using ELISA,HE staining and Western blotting(Western blot,WB)technology,to observe the Protein expression and CIA of rat knee joint morPhology and IL-17,IL-23,VEGF content,to exPlore the mechanism of electroacuPuncture to treatment of rheumatoid arthritis.Methods 60 male Wistar rats were randomly taken 8 as normal control grouP,and the rest used infreund' s adjuvant and tyPe ? collagen to induce arthritis.24 rats which successf ? lly induced were randomly divided into three grouPs:model,Prednisolone grouP and electro-acuPuncture(EA)grouP,8 in eaeh grouP.Treatment from the 21th day,the normal grouP and model grouP was given saline solution(0.1 ml/10 g)to fill the stomach,and Prednisone grouP was given Prednisone solution(0.1 ml/10 g)to fill the stomach.The EA(2/100Hz,1 2mA)grouP was aPPlied to "Zusanli”(ST36)and "Kunlun"(BL60)for 30 min,once daily for 10 times.To measure the rats' body weight and ankle diameter before and after the building,and the end of the treatment.Then Plasma VEGF,IL-17 and IL-23 contents in serum and the knee joint cavity by ELISA,immunohistochemical method and Western blot.Results 1.General:ComPared with the normal grouP,the rest of the grouP rats,food volume and weight have different degrees of reduction,Paw swelling,ankle swelling and arthritis score index more than eight Points,and the knee joint Pathological bioPsy show local inflammatory cell infiltration.all of these can indicate that building success.ComPared with model grouP,after the treatment by EA and Prednisone,the symPtoms can imProve.ComPared with Prednisone,EA is suPerior to Prednisone in terms of increasing body weight,but the rest were no comparative difference(P>0.05).2.weight:the weight of the rats during the exPerimental Period is uPward.ComPared with the control grouP,the weight of modeling rats increased slowly.After EA or Prednisone treatment,the trend is increasd.3.AI:electro acuPuncture can relieve joint swelling,low AI.4.Observing the rat knee section found that the surface of the normal grouP rats joint is smooth and no inflammatory cells infiltration.Model grouP rats cartilage surface is uneven,having a large number of inflammatory cells infiltration,especially tibia dried scale end.The rats treated by electric acuPuncture or Prednisone,artic?lar surface is smooth,joint sPace narrow,and a small amount of inflammatory cells infiltration.5.Protein content:By ELISA method and Protein imPrinting method to detect the IL-17,IL-23 and VEGF levels in the knee joint and serum show that comPared with normal grouP,the model grouP rats are significantly higher in the level of IL-17,IL-23 and VEGF(P<0.05);while EA grouP and Prednisone grouP are significantly lower than the model grouP;compared with EA grouP and Prednisone grouP,in addition to WB is slightly higher,the rest were no statistical difference(P>0.05).Conclusion(1)EA can imProve the CIA rats eating and joint swelling degree significantly,then has obvious theraPeutic effect on CIA rats.(2)Electric acuPuncture can relieve inflammatory cells infiltration,and imProve Pathological state of knee joint.(3)EA can inhibit the Over expression of knee joint tissue IL-17,IL-23 and VEGF Protein content,blocking the IL-23/IL-17 axis,inhibiting angiogenesis,bone destruction.This may be one of the imPortant mechanisms of acuPuncture treatment of RA.
