Part ?Experimental study about targeted anti-gastric cancer in nude mice by autocrine of TNF-? oncolytic adenovirus using cik cells as vehiclesObjective The comprehensive treatment at present is not satisfactory if discovering recurrence and metastasis lesions of gastric cancer.Exogenous TNF-? is easily eliminated by the body in the process of killing gastric cancer and can not reach a perfect outcome.This project is expected to solve the problem of anti-metastasis and recurrence of gastric cancer.Method This project uses automatic bead extraction and purification system to isolate CIK cells from normal human peripheral blood,and transforms the oncolytic virus to carry TNF-? gene through the technology of molecular genetic engineering.This experiment establishes animal models of gastric cancer bearing nude mice,and is designed to study transplantation tumor volume of gastric cancer nude mice model,the expression of TNF-? in cik cells and clinicopathological necrosis features of gastric cancer tissue.Result CIK cells as the carrier carry recombinant oncolytic adenovirus which bring the TNF-? can be targeted to reach the gastric cancer tissues based on the cells targeting to gastric cancer.CIK cells which kill gastric cancer cells by direct and indirect immune response can release TNF-? oncolytic adenovirus after their own apoptosis.Conclusion AD-TNF-?-EGFP CIK cells showed strong anti-tumor effects on gastric cancer by proliferation,autocrine and releasing TNF-? after adenovirus infected tumor cells.Part ? Expression and clinical significance of CD90 in gastric cancerObjective The expression level of CD90 and mononuclear macrophage in gastric cancer tissue,the tissue samples adjacent to carcinoma and the chronic gastritis was measured by immunohistochemistry and their relationship with clinicopathological parameters and prognosis was analyzedto investigate the expression and significance of CD90 in gastric cancer tissue.Method The 96 gastric cancer tissue specimens with corresponding normal epithelium tissues were selected from Qingdao municipal hospital general surgery database from January 2006 to June 2008,and 20 cases of chronic atrophic gastritis group were derived from the gastroscope biopsy specimens.The clinical data and follow-up data were completed by phone or outpatientuntil January 2014.We collected the case data of the gastric cancer patients were included age,sex,site,pathological type,TNM staging and so on.Statistical analysis was performed through SPSS software 17.0.The expression level of CD90 and the clinicopathological characteristics were examined by using Pearson chi-square.Survival curve was evaluated by using the Kaplan-Meier method,and we adopt Cox regression analysis to multivariate survival analysis.All analyses were considered to be statistical significant when p values <0.05.Result(1)High expression of CD90 in 66.7%(64/96)gastric cancer tissue samples,compared with 15.6%(15/96)normal tissues and 15.0%(3/20)chronic gastritistissues(p<0.01).(2)High expression of CD90 was significantly associated with advanced TNM stage(P=0.014),p T(P=0.02),p N(P=0.017)and p M status(P=0.045),and irrelevant with age,gender,pathological type and parts.(3)Furthermore,patients with high expression of CD90 had worse overall survival for 5-year(P<0.05 in multivariate analysis).Conclusion CD90 may have correlation with invasiveness and metastasis of gastric cancer.And CD90 might serve as a new molecular targets for therapy,and judging the prognosis of patients with gastric cancer.
