Objective: To investigate the differences of CD90 and CD44 expression in human hepatocellular carcinoma(HCC)and distal normal liver tissues,and to elucidate the differences and clinical significance of CD90 and CD44 in hepatocellular carcinoma and normal liver tissues.Method:In the experimental group,40 cases of human hepatocellular carcinoma tissues were collected from the Affiliated Tumor Hospital of Guangxi Medical University from December 2013 to December 2014.And the corresponding distal liver tissue samples were taken as the control group,the clinical datas associated with each patient were also recorded.The expressions of CD90 and CD44 mRNA were detected by real-time quantitative PCR(FQ-RT-PCR).The expressions of CD90 and CD44 protein were detected by immunohistochemistry.Result: All tissues were confirmed by pathologic observation again.The results showed that the experimental group was changed in hepatocellular carcinoma: the cell shape was irregular,the nucleus was larger,the nuclei was stained differently,the proportion of nucleus and nucleus increased,and the pathological kyphosis was found.The control group was normal liver cell morphology.The results of RT-PCR showed that the expression of CD90 mRNA in human hepatocellular carcinoma was(10.210 ± 1.469)higher than that in distal normal liver(9.329 ± 1.760),the difference was statistically significant(P<0.05).The expression of CD44 mRNA in liver cancer tissues(9.269 ± 1.328)was higher than that in distal normal liver tissues(7.775 ± 1.526),the difference was statistically significant(P <0.05).The results of immunohistochemical staining showed that the expression rates of CD90 and CD44 in hepatocellular carcinoma were 82.5%(33/40)and77.5%(31/40)respectively,and in distal normal liver tissues were 17.5%(7/40)and 12.5%(5/40)respectively.The expression of CD90 and CD44 in hepatocellular carcinoma was higher than that in distal normal liver,the difference was statistically significant(P <0.05);Among them,positive staining of CD90 antigen was mainly located in cytoplasm,CD44 molecular antigens are highly expressed in the cell membrane of hepatocarcinoma cells,brownish and cytoplasmic.CD90 molecular antigen staining mainly located in the cytoplasm,CD44 molecular antigen located in the cell membrane of liver cancer cells,cytoplasm is also expressed.The expression of CD90 and CD44 did not correlate with the age,sex,alpha-fetoprotein(AFP)and tumor size of the patients,CD90 and CD44 molecules are associated with the invasion,pathological grade,clinical stage and portal vein thrombosis.CD90 and CD44 were not associated with survival rates in patients with liver cancer,were not associated with survival.Conclusion:1.The expression of CD90 and CD44 in hepatocellular carcinoma was significantly higher than that in normal control group.2.The results of this study show that the expression of CD90 and CD44 is related to the invasion,pathological grade,clinical stage and portal vein thrombosis,is notassociated with survival. |
