| Grassland ecosystem belongs to terrestrial ecosystem,which is rich in biodiversity and plays an important role in animal husbandry production.Therefore,maintaining grassland ecosystem balance is of great significance to animal husbandry development.Rodents are one of the main factors affecting the balance of grassland ecosystems.It not only destroys the balance of grassland ecosystem,but also causes great economic losses to agriculture and animal husbandry in China.Brandt’s vole is the dominant species of Inner Mongolia grassland,and when it is too large,it will cause serious damage to grassland and grassland ecosystem.There are many factors that affect the number of animal populations.Food abundance is one of the main environmental factors in grassland ecosystem.Does the abundance of food affect the population of grassland ecosystems and whether the genetic diversity of grassland populations,the impact of such issues have not been reported.In this study,the control of the environmental factors in Inner Mongolia grassland animal experimental research station and the food treatment and control of the fence in the Brandt’s vole population as a material,the genetic diversity of Brandt’s voles was detected by using 10 polymorphic microspot-rich microspheres,effects of food abundance on Genetic Structure of Brandt ’s Voles Population.In this experiment,the genomic DNA of Brandt’s vole was extracted by the conventional phenol / chloroform extraction method,and the extracted genomic DNA was detected by agarose gel electrophoresis,(FAM or HEX)primers were used for PCR amplification,ROX-500 was used as the internal standard for molecular weight,capillary electrophoresis was performed using a 3730 XL sequencer,genotyping of the detected microsatellite loci,the number of alleles(Na),the number of effective alleles(Ne),observation of heterozygosity(Ho),expected heterozygosity(He),polymorphic information content(PIC),Shannon’s information index and other genetic diversity indicators were measured by Popgene software,PIC-CALC software,GenAlEx 6.502 and MEGA4x1 software for statistical analysis.From the statistical results we can see,the average number of alleles of the feeding fence and control fence are respectively 8 and 5.3,the average effective allele numbers are respectively 4.7677 and 3.7943,the average observed heterozygosity are respectively 0.71 and 0.6167,the average expected heterozygosity are respectively 0.7789 and 0.6871.The difference between the indicators are significant analysis that the genetic diversity of the feeding fence are significantly higher than that of the control fence(P<0.05).The results of this study show that food abundance affects the genetic structure of animal populations and regulates the number of animal populations.The results can help to further understand the genetic control mechanism of the fluctuation of animal population quantity,and provide scientific basis for the comprehensive prevention and control of grassland rodents and the protection of grassland and grassland ecosystem. |
