The abiotic stresses such as salt,drought,high temperature or low temperature seriously affected the sustainable development of modern agriculture.Plants could improve the ability of resistance to the stresses by regulating the abundance and stability of RNA.NTP(Nucleotidyl transferase protein)contained poly(A)polymerase domain and NT domain which could add non-templated nucleotides to the terminal of RNA,changing the abundance and stability of RNA.Although preliminary studies have been reported about some NTPs in plants,but most functions of NTPs still remained unclear.In this study,we used bioinformatics to analyze the protein sequence and structure of 10 NTPs in Arabidopsis thaliana.At NTPs had the same conserved domain but were different in acid-base properties.NTPs could interact with other proteins.For example,10 At NTPs interacted directly with CSDP1,which was encoded by a stress-related gene.CSDP1 could response to stresses directly.All of the At NTPs had one type or more stress-related cis-acting elements in their promoters,which implied that the NTPs might be involved in some stress responsing pathways.This result was validated by the real-time PCR analysis of the expressions of At NTP genes under salt,drought,cold and heat treatments.We found that HESO1,NTP6 and NTP7 were induced by salt,drought,cold and heat treatments.The physiological indexes of heso1,ntp6 and ntp7 under salt stress had been observed.Compared with Col-WT,the germination rate,the root length and the green cotyledon were lower in heso1,ntp6 and ntp7.The tissues of heso1,ntp6 and ntp7 were seriously damaged under drought stress,which indicatd that the resistance ability of the mutations of HESO1,NTP6 and NTP7 were decreased under drought treatment.By analyzing the RNA-seq data of three ntp mutants under the salt stress ?,we found that many genes such as the key gene in response to stresses-KIN1 changed in the mutations of NTP gene.In conclusion,we found that HESO1,NTP6 and NTP7 were induced by abiotic stresses and affecting the stress tolerance of Arabidopsis thaliana,indicating that HESO1,NTP6 and NTP7 might be involved in the process of plant response to the environmental stresses.
