The growing demand for public health in the world, paves the way for the development of biochemical sensors.Sensing technologies can be divided into two categories according to the detection method: label-based detection and label-free detection. The biochemical sensor according to the signal conversion mechanism can be divided into four categories: electrical, mechanical, magnetic and optical biochemical sensors. For a number of years, the scientific community has been paying growing attention to label-free optical biosensors due to its combination of advantages of label-free technology(low assay costs and complexity, reusable)and of optical biosensors(electromagnetic immunity, less invasive, high sensitivity). In this paper,an optical microfluidic sensor based on Fabry-Perot resonator is designed and fabricated. It has the characteristics of low cost, compact structure, wide detection range and low detection limit.Metal film is fabricated into Fabry-Perot resonator. The detection performance of the sensor on biological samples is obtained. In addition,a compact stable test system with low noise is designed and built. The sensor is made of metal coated glass and has good temperature stability. When the functional layer covered on the metal film captures the target molecule, the optical path of light beam in the optical cavity changes. Thus a detection of concentration range of 10-10 g?m L-1-10-4g?m L-1 of thrombin solution is achieved.Glutathione is also tested as an example of small molecule.Feasibility is showed that detection of gold-sulfur bond with the biosensor, demonstrating that the sensor can function in a variety of applications.
