As the microfluidic chip's micromation of structure and high specific surface area, the physical and chemical properties on surface play an important role in microfluidic analysis. In the scale of micrometers, the surface effect of microchannel wall has great influences on the flow inside microchannel,the shape and size of inner wall of micro channel, the surface roughness, hydrophilic and other physical properties are the important factors effecting microfluidic chip,which will affect the PCR amplification directly. In order to improve the performance of microfluidic chip and improve the physical and chemical properties of inner surface of microchannel,the modification on microchannel inner-surface has become one of the research focuses in the field of microfludic control.After the paper investigation of overseas and domestic literatures, this paper based on modification of microchannel, and also do some research related to the application of key experimental techniques after modification.Combined with the condition of laboratory, the main work did as follows:(1)By invested lots of literatures aout the microfluidic chip within microfluidic field, leads to the research background, some related key technologies and principle were described, and summarized the domestic and foreign research status, then the advantages and disadvantages of the existing research are commented.(2)Modified microfluidic chip in a new method with using UV cured materials,which is based on air blown way through UV curing optical adhesive NOA68 to improve microfluidic chip, a double microfluidic chip were modified in order to reflect advantages of this new method. The method achieved good results by the comparion of the roughness and hydrophilicity of inner microchannel wall before and after modification.By PCR experiments, the efficiency of PCR amplification is improved after the modification.(3)In order to ensure the smooth progress of the PCR experiment when the experiments was carried out on the modified microfluidic chip, some key experimental techniques are studied and designed in this paper. The continuous sampling device for microfluidic chip are designed, the temperature on the surface offilm heater was basically uniform by simulation, the hardware circuit and software program are also designed, finally, the temperature fluctuated 0.9?.(4) The endpoint fluorescence detecter are made that suit for one sample detection. While,this simple detection method has some limitations.At last, according to the special structure of the double layer microfluidic chip, real-time fluorescence signal detection device which has possession of intellectual property are designed in this paper.
