| Objective: To construct tissue microarray of gastric cancer and investigate the expressions and correlations of gastrin and apoptosis-associated proteins involved in mitochondrial apoptotic pathway in gastric cancer tissues,and explore their clinicopathological characteristics.Methods: The tissue chip technology and Evision of immunohistochemistry(IHC)were used to detect the expressions of gastrin and apoptosis associated proteins involved in mitochondrial(Bcl2、BAX、caspase-9 and caspase-3)in human gastric cancer tissues and their paracancerous tissues.The correlation of these markers and their clinicopathological characteristics were analyzed using Spearman rank correlation analysis and chi-square test.Results: 12 tissue microarrays(TMAs)are established successfully.The expressions of gastrin and Bcl2 in gastric cancer were significantly higher than those in the corresponding paracancerous tissues(P<0.01),whereas the expressions of caspase-9 and caspase-3 in gastric cancer tissues was significantly lower than those in the corresponding tissues(P<0.05).There was no significant difference in the expression of BAX between gastric cancer tissues and the corresponding tissues.There was a significant positive correlation between the expressions of gastrin and Bcl2 in gastric cancer tissues(r=0.237,P<0.05).The expression of gastrin was associated with tumor position(P<0.01),which was significantly higher in the antrum and corpus of gastric than that in the gastric cardia.The expression of Bcl2 was associated with tumor size(P<0.05),TNM stage(P<0.05)and lymph node metastasis(P<0.01),which was higher in gastric cancer with tumor diameter less than 5cm and the negative lymph node metastasis and in TNM I stage than that with tumor diameter more than 5cm and the positive lymph node metastasis and in TNM II、III、IV stage.The expression of caspase-9 was associated with age(P<0.05).Conclusion: Gastrin and Bcl2 are highly expressed in gastric cancer tissues,and they are correlated with the clinicopathologic features. |
