| Background Psoriasis (OMIM *177900) is a common, chronic and recurrent, inflammatory skin disease with a prevalence of 0.123% in Chinese Han population. The pathogenesis of psoriasis remains unclear. Now most people tend to think that genetic factors,immunology factors and environmental factors play important roles in the pathogenesis of psoriasis. Psoriasis vulgaris (PV) is the most frequent clinical type, more than 90 percents. Recently, we found a distinguished correlation of rs2431697 polymorphism in PTTG1 gene with psoriasis vulgaris in Chinese Han Population by psoriasis genome-wide association study (GWAS).Objective To investigate the potential association of PTTG1 gene polymorphism and specific phenotypes of psoriasis vulgaris in Chinese Han Population basing on stratifying analysis of the age of onset, family history and clinical types, and to provide an important genetic basis in exploring the pathogenesis of psoriasis.Methods We analyzed the distributions of genotype and allele of rs2431697 (polymorphic site within PTTG1 gene) in 5,468 cases and 6,406 normal controls, which derived from the psoriasis GWAS carried out by our group. The appropriate application of social science statistical software package SPSS 10.0 was used for statistical analysis of data. Chi square test was used for comparing genotypic or allelic frequencies distribution among the groups.Results 1. The distributions of alleles and genotypes for rs2431697 were significant differences between cases and controls (P=3.20×10-9,P=3.10×10-8, respectively). Then cases were stratified from age of onset, family history and clinical type, there were also significant differences between clinical subgroups and controls in distributions of alleles and genotypes (all P<1.0×10-3). 2. The distribution of alleles for rs2431697 was significantly different between familial case and sporadic case groups (χ2=5.47, P=0.02),while there was no significant differences between familial case and sporadic case groups in the distribution of genotypes for rs2431697 (χ2=5.41, P=0.07). 3. There were no significant differences between the pediatric onset psoriasis patients and the adult onset psoriasis patients in distribution of alleles and genotypes of rs2431697 (χ2=0.93,P=0.34;χ2=0.92,P=0.63; respectively). 4. No significant differences were found in distributions of alleles and genotypes for rs2431697 between the guttate and plaque cases (χ2=0.09,P=0.76;χ2=0.09,P=0.96; respectively).Conclusion 1. The polymorphism of PTTG1 gene (rs2431697) is associated with the susceptibility of psoriasis vulgaris in the Chinese Han population; 2. The polymorphism of PTTG1 gene (rs2431697) is associated with family history of psoriasis vulgaris; 3. The polymorphism of PTTG1 gene (rs2431697) may not be associated with age of onset of psoriasis vulgaris; 4.
