Objective The aim of this study is to observe the level of DNA methylation of peripheral blood thymocyte differentiation antigen-1 and the expression of methyltransferase and methyl-binding protein 2 in pneumoconiosis from the perspective of epigenetics,and to achieve the goal of analyzing the relationship between DNA methylation and dust-induced pulmonary fibrosis.In this paper,in order to meet the purpose of this study,the auther also observed the changes of the expression of COL-I,COL-III and TGF-?1,Smad2/3 and TNF-?in serum in pneumoconiosis,and further explored the role of Thy-1 DNA methylation in the pathogenesis of pneumoconiosis to find an effective early diagnosis target for pneumoconiosis.Method In this study,84 cases of pneumoconiosis diagnosed by Occupational Disease Prevention and Control Center of the X of Shizuishan City in Ningxia were selected as the pneumoconiosis group and 75 healthy coal miners workedin the same place with the corresponding exposure history were served as the control group.The methylation levels of Thy-1 were detected by nM-PCR,the expressions of DNMT1,DNMT3A,DNMT3B and MeCP2 were detected by qRT-PCR,the expressions of COL-I,COL-III,Smad2/3?TGF-?1?TNF-?were detected by ELISA.Results1.The average age and length of service in the control group are?43.80±6.63?years old and?23.35±7.92?years.The average age and length of service in the pneumoconiosis group are?48.30±5.05?years old and?27.49±5.04?years.In the pneumoconiosis group,the coal mining workers account for 45.2%,boring worker is 42.8%and helper is 12.0%.2.The levels of COL-I and COL-III in serum of pneumoconiosis group are higher than those in control group?P<0.05?.The levels of COL-I and COL-III in serum of silicotic group are higher than those in control group and coal miners pneumoconiosis group?P<0.05?.The level of COL-I in stage I and stage II silicosis group are higher than those in control group and stage I coal miners pneumoconiosis group?P<0.05?.The level of COL-III in stage I and stage II silicotic group are significantly higher than those in control group?P<0.05?.3.The levels of TGF-?1 and Smad2/3 in pneumoconiosis group are higher than those in control group?P<0.05?.The level of TGF-?1 in coal miners pneumoconiosis group is higher than that in control group?P<0.05?.The levels of TGF-?1 and Smad2/3 in silicotic group are pneumoconiosis and stage I silicosis?P<0.05?.7.Correlation analysis:There are a positive correlation between the stage of coal worker's pneumoconiosis and the expression of COL-I,TGF-?1,Smad2/3 and TNF-??P<0.05?;There are a positive correlation between the stage of silicosis and the level of Thy-1 methylation and the expression of COL-I,COL-III,TGF-?1,Smad2/3 and TNF-??P<0.05?;Thy-1methylation is positively correlated with the expression of TGF-?1,Smad2/3 and TNF-??P<0.05?.The working age is positively correlated with the expression of TGF-?1,Smad2/3 and TNF-??P<0.05?,and the work is positively correlated with the expression of TGF-?1 and TNF-??P<0.05?.Conclusion Under the action of DNMT3B and MeCP2,Thy-1 DNA hypermethylation occurs in the peripheral blood of patients with pneumoconiosis,which might be an important epigenetic mechanism during the development and progression of pneumoconiosis.
