OBJECTIVE: This study was to investigate the changes in the expression of HFABP,Tn-T,caspase3,and RIP3 after myocardial infarction to confirm whether the combined detection of these factors is valuable for the diagnosis of acute myocardial infarction.METHODS: This study was divided into two parts.The first part was a rat model of myocardial infarction with ligation of the left anterior descending artery.The rats was divided into eight groups :(1)the control group,(2)the sham operation group,(3)15min group,(4)30min group,(5)1h group,(6)3h group(7)6h group and(8)9h group.We used ECG to judge the success of the model.After myocardial tissue was drawn,TTC and HE were stained in the heart of rats.We used immunohistochemistry and immunofluorescence techniques to observe the expression of HFABP,Tn-T,caspase3,and RIP3 in myocardium after acute myocardial infarction.Western blotting was used to quantitatively analyze the expression of HFABP,Tn-T,caspase3,and RIP3.ELISA was used to analyze the blood HFABP and Tn-T levels after myocardial infarction in rats.And we used electron microscopy to observe the microscopic changes of myocardial morphology after myocardial infarction.In the second part,We selected 30 myocardial tissue specimens from Department of Pathology,NingXia Medical University from 2007 to 2016.These specimens are divided into three groups:(1)the control group,(2)the coronary artery stenosis group,and(3)the myocardial infarction group.We performed HE staining on selected specimens to observe coronary stenosis and its corresponding myocardial changes.Immunohistochemical staining was used to observe the expression changes of HFABP,Tn-T,caspase3,and RIP3 in myocardium.RESULTS: The first part of the experiment,(1)after the successful operation,the electrocardiogram showed that S-T segment arch-like elevation,(2)TTC staining showed that infarcted myocardium pale,non-infarcted myocardium red,(3)HE staining showed that after myocardial ischemia,the myocardial cells were arranged disorderly,with cytoplasmic edema,light staining,disappearance of transverse striations in the myocardium,nuclear condensation,deep staining,and focal infiltration of inflammatory cells and we also found that the wavy changes in the heart muscle.(4)Immunohistochemistry and immunofluorescence staining showed that after 15 minutes of ligation of the left anterior descending branch of the coronary artery,HFABP and Tn-T in the ischemic myocardium could show patchy loss,with the prolongation of ischemic time,the HPABP and Tn-T were obviously missing,but the expression of caspase3 and RIP3 increased.(p<0.05 vs.control group).(5)Western-blotting detection of myocardial HFABP,Tn-T,caspase3 and RIP3 showed that with the prolonged ischemia time,HFABP,Tn-T content decreased,caspase3,RIP3 content increased(p<0.05 vs.control group).(6)Using of ELISA technique to detect HFABP and Tn-T levels in rat's blood after myocardial infarction indicated that HFABP and Tn-T in blood increased with the prolongation of ischemic time.(p<0.05 vs.control group).(7)Electron microscopy showed the microscopic changes of myocardial morphology after myocardial infarction that mitochondria were loose,degenerate,swollen,and mitochondrial hemorrhoids dissolved and fractured.In the second part of the experiment,(1)HE staining showed that the coronary stenosis was visible in the coronary artery stenosis group and the myocardial infarction group,and a large number of neutrophils were seen in the myocardial infarction group.(2)Immunohistochemical staining showed that HFABP and Tn-T were strongly expressed in the control group,the expression in the coronary artery stenosis group and myocardial infarction group was weak or absent,the expression of caspase3 and RIP3 was weak in the control group,and in the coronary artery stenosis group and myocardial infarction group the expression enhancement(p<0.05).CONCLUSIONS: HFABP,Tn-T,caspase3,and RIP3 expression changes in early acute myocardial infarction.Combined detection of these factors has a certain value in the early diagnosis of acute myocardial infarction.
