Objective:To study whether Saussurea tridactyla Sch.-Bip.-derived flavones have protective effect after oxidative damage in UVB-irradiated HaCaT cells.Method:HaCaT cells cultured in vitro were randomly divided into 2 groups,which received30mJ/cm~2 and 60mJ/cm~2 UVB.The 2 groups subdivided into 4 groups,includinga control group,UVB group,Saussurea tridactyla Sch.-Bip.-derived flavones 2 g/L group 6 g/L group.Cells in the flavones groups were were treated with 2g/L and 6g/L flavones before UVB irradiation,respectively.Cells were collected after UVB irradiation at different doses of UVB for 1h,then cultivated for18h.The morphology of cells were observed by by trypan blue staining;cells viability were detected by MTT.The SOD activity,GSH content,MDA content,and CAT activity of HaCaT cells were evaluated by using SOD kit,GSH kit,MDA kit,and CAT kit.Elisa was used to detect the levels of P38MAPK,p53,Bax and Caspase-3 and Bcl-2.Results:In low and high dose of UVB groups,the number of blue cells were increased in UVB group when compared with the control group.The cells were floating and swelling,the morphology of some cells were blur;the absorbance of each group showed a downward trend(P<0.05);the SOD activity,CAT activity and GSH content in the UVB group were significantly lower than that in the control group,and the content of MDA significantly higher(P<0.05).And the levels of P38MAPK?P53?Bax?Caspase-3 were increased while the Bcl-2 level was decreased(P<0.05).In flavones groups,the number of blue cells were decreased,cell edema and apoptosis were alleviated;the absorbance showed an upward trend(P<0.05);SOD activity,CAT activity and GSH content were significantly higher and the content of MDA significantly lower(P<0.05).And the levels of P38MAPK?P53?Bax?Caspase-3 were decreased while the Bcl-2 level was increased(P<0.05).Conclusion:Saussurea tridactyla Sch.-Bip.-derived flavones may alleviate oxidative damage in UVB-irradiated HaCaT cells and photoaging.
