Objective A model of skin aging in vivo and in vitro established by ultraviolet(UVB)radiation and subcutaneous injection of D-galactose was established.Oral intervention with Saussurea Medusa Maxim Polysaccharides was performed to observe its effects on the structure of mouse skin aging model and oxidative stress indicators.Methods 50 SPF mice were divided into five following groups: normal control group(Control group),model group(UVB group),vitamin E group(UVB + VE group),and Saussurea Medusa Maxim Polysaccharides low-dose group(LSMMP group).Saussurea Medusa Maxim Polysaccharides high-dose group(HSMMP group),10 in each group.Except the Control group,the skin of the neck and back of mice in the other groups was subjected to daily UVB irradiation and subcutaneous injection of 5% D-galactose at 10 ml / kg.From the 11 th day of the experiment,the control group and the UVB group were given an equal volume of normal saline,the UVB + VE group was given a 1 g / kg / d vitamin E solution,the LSMMP group was given 1 g / kg / d,and the HSMMP was given 4 g / kg / d.Observe the skin changes on the back of mice;HE staining and Masson staining to observe pathological changes;GSH-Px,SOD activity,and MDA,Hyp levels were measured by enzyme biochemical method.Results Except the Control group,skin photoaging changes occurred in all groups except the control group.Among the UVB group,obvious erythema,scaling,static deep wrinkles,etc.,were found on the skin of the neck and back of the mice.However,group HSMMP was definitely close to the Control group.Under the light microscope,the epidermis thickened obviously,the dermal elastic fibers were broken,curled,and the number of collagen fibers was obvious,with irregular arrangement.The VE + UVB,LMSSP,and HSMMP groups alleviated the symptoms of photoaging,and the HSMMP was closer to the Control group.Compared with the Control group,the SOD activity,GSH-Px activity,and Hyp content of the UVB group were significantly reduced,and the MDA content was increased(P<0.05).Compared with the UVB group,the SOD activity,VE + UVB,LMSSP,and HSMMP groups,GSH-Px activity and Hyp content increased significantly,and MDA content decreased(P <0.05);there were differences between the three groups(P <0.05).Conclusion The Saussurea Medusa Maxim Polysaccharides have a protective effect on mouse skin aging model,reduces oxidative damage and delays skin aging.
