Objective:To explore the level of expression of LncRNA-Lucat1 in clear cell renal cell carcinoma(ccRCC)and analyze the relationship between Lucat1,expression and prognosis of ccRCC patients.And to investigate the effects of LncRNA-Lucat1 on proliferation and metastasis of ccRCC cell lines.Methods:At the beginning,we searched TCGA database to analyze the expression of LncRNA-Lucat1 in tumor tissues and para-tumor tissues of ccRCC and figure out the relationship between Lucat1 expression and clinical pathological parameters of ccRCC.Survival curve was plotted by the Kaplan-Meier method and compared by the log-rank test.Secondly,to verify the outcome of TCGA,qRT-PCR was used to detect th expression level of Lucat1 in 45 pairs Clinical specimens and 4 ccRCC cell clones.Lastly,we constructed expression vector of shRNA to validate the effection of Lucat1 on the biological property of ccRCC cell lines.Results:1.Through TCGA data base we found that the expression of LncRNA-Lucat1 in ccRCC tumor tissues is relatively higher(2.14 times,P < 0.01).The expression level of Lucat1 has not correlation with clinical pathology parameters such as gender(P = 0.115),age(P = 0.108)and recurrence(P = 0.225).However,some parameters like pathological grading(P=0.015),TNM stage(P=0.001)and metastasis(P=0.009)had a vital interrelation with the expression of Lucat1.We discovered that high Lucat1 expression in ccHCC tissues was associated with significantly worse survival of ccRCC patients(P< 0.001).2.Lucat1 Knockdown inhibited ccRCC cells proliferation,migration and invasiveness in vitro(P< 0.001).Conclusion:Lucat1 prompts ccRCC cell proliferation,migration and invasiveness and it may act as a role of oncogene in the occurrence and development of ccRCC.
