| Gut immunity is a defense system against external harmful substances in Drosophila melanogaster.The project aims to explore the mechanism of Rheum officinale extract on gut immunity.The survival rate of D.melanogaster was measured after treated with Paraquat,Sodium dodecyl sulphate(SDS)and Dextran Sodium Sulfate(DSS).The gut injury model was constructed by SDS.The numbers and size of progenitor cells in intestinal epithelium cells,the amount of intestinal stem cells(ISC)in dividing,the numbers of enterocytes and enteroendocrine,the activity of JNK signal pathway and the levels of reactive oxygen species and dead cells were detected by fluorescence immunstaining.And the gut morphology and lifespan of D.melanogaster were also analyzed.The proliferation and differentiation of intestinal stem cells were regulated by a variety of factors.In order to explore the effect of CG7510 gene on the proliferation and differentiation of intestinal stem cells.The survival dates was observed by feeding with Paraquat,DSS and Gram-negative bacterium,Erwinia carotovora carotovora 15(Ecc15).The numbers of progenitor cells and the dividing intestinal stem cells(ISC)were detected in the normal or gut infection condition.And the numbers of intestinal stem cells,enteroblast,enterocytes and enteroendocrine were also detected.The results showed that Rheum officinale extract could significantly improve the survival rate of D.melanogaster after treaded with Paraquat,SDS and DSS,regulate the activity of JNK signal pathway,alleviate excessive proliferation and differentiation of intestinal stem cells induced by injury.What’s more,Rheum officinale extract could reduce the numbers of dead cells and the levels of reactive oxygen species(ROS),maintain the gut morphology and improve the lifespan of D.melanogaster.The results demonstrate that Rheum officinale extract can alleviate gut damage of D.melanogaster caused by external substances and enhance the gut immune function.The results showed that low expression of CG7510 in progenitor cells resulted in a decrease in Drosophila survival and a decrease in the number of progenitor cells under gut infection.Under normal condition,the phenomenon of overproliferation of intestinal stem cells and excessive differentiation of precursors in CG7510 low expression mutants were detected.The results show that CG7510 plays an important role in the proliferation and differentiation of intestinal stem cells.This finding provides an important theoretical basis for the study of the function of genes in gut immunity... |
