Objective:To evaluate the effect of Peiminine on Ventilator induced lung injury in mice,and compare the effect with dexamethasone.Methods:Thirty-two male BALB/c healthy mice made a contribition to the study which were divided into four groups randomly(n=8);The Control group(group C)and model group(group V)were given normal saline(NS)for 7 days by intragastric administration;Dexamethasone group(group D)was given dexamethasone for 7 days by intragastric administration;Peiminine group(group P)was given peiminine for 7 days by intragastric administration;Blood sample and lung tissue were obtained after intragastric administration in group C,while in group V,group D,group P,blood sample and lung tissue were extracted after 4 hours of mechanical ventilation on the 7th day of intragastric administration.The blood levels of interleukin-8(IL-8),tumor necrosis factor-?(TNF-?)and lung tissue levels of aquaporin 1 and 5 were measured with ELISA;left lung was stained with hematoxylin and eosin(H&E)for pathological examination.Results:1.Compared with group C,the levels of TNF-? and IL-8 in group V was increased(P<0.05),AQP-1 and AQP-5 of group V were decreased(P<0.05),tissue Pathological damage of lung tissue was obvious in group V;there was no significant difference in TNF-?,IL-8,AQP-1 and AQP-5 contents between group D?P and C;The pathological damage of lung tissue in group D and P was more severe than group C.2.Compared with group V,the levels of TNF-? and IL-8 in group D and P were decreased(P<0.05),AQP-1 contents increased significantly(P<0.05),AQP-5 contents was not statistically significant(P>0.05),Pathological damage of lung tissue was lighter in group D and P.3.Compared with group D,there was no significant difference in the contents of TNF-?,IL-8,AQP-1 and AQP-5 in group P(P>0.05),Pathological damage of lung tissue was similar to group D.Conclusion:Peiminine pretreatment can reduce ventilator induced lung injury in mice;the effect is similar to dexamethasone.Its possible mechanism of action is reducing lung tissue inflammatory factor and increase lung tissue AQP-1 contents.
