Objective: Luteolin is one kind of flavonoid found in many plants or herbs.At present,there are lots of studies on pharmacological its activities,especially on its relationship between luteolin and cancer.However,the inhibitory effect of luteolin on human tongue squamous carcinoma Tca8113 cells has not been reported until now.Our study investigated the effects of luteolin on the proliferation,apoptosis and metastasis of Tca8113 cells and explored its preliminary mechanisms.Methods: The sulforhodamine B(SRB)method was used to detect the proliferation viabilities of human tongue squamous carcinoma Tca8113 cells,liver cancer HepG2 cells and human microvascular endothelial HMEC-1 cells.DAPI nuclear staining and flow cytometric method were done to determine the effect of Tca8113 cell apoptosis.The invasion and migration experiment through Transwell chamber was adopted to detect the migration and invasion effects of Tca8113 cells.The adhesion experiment through matrigel was conducted to assay the adhesion effect of Tca8113 cells.The MMP-2 and MMP-9 expressions in Tca8113 cells were assessed through Gelatin Zymography assay.Western blotting assay was done to determine protein expressions of Akt,p-Akt,IKK and p-IKK in Tca8113 cells.NF-?B nuclear transfer capacity in Tca8113 cells was done by immunohistochemistry assay.Results: Our results showed that luteolin inhibited the proliferation viabilities of Tca8113 cells,HepG2 cells and HMEC-1 cells,and the inhibitory effect on Tca8113 cells was the most potent in a concentration-dependent and time-dependent manner.It also concentration-dependently induced Tca8113 cell apoptosis,inhibited the migration,invasion and adhesion of Tca8113 cells,and reduced the expression of MMP-2 and MMP-9 in Tca8113 cells.Moreover,luteolin inhibited the protein expression of Akt,p-Akt,IKK and p-IKK and the nuclear transfer ability of the transcription factor NF-?B in Tca8113 cells in a concentration-dependent manner.Conclusion: Luteolin can inhibit the proliferation of human tongue squamous carcinoma Tca8113 cells and induce Tca8113 cell apoptosis.It also effectively inhibitsthe invasion,migration,and adhesion of Tca8113 cells,and decreases the expression of MMP-2 and MMP-9,which suggests that luteolin can inhibit the metastasis of Tca8113 cells.Its mechanism may be related to the PI3K-Akt-NF-?B signaling pathway.
