Study On Quality Control Method And Purification Technology Of Anti-inflammatory Effective Parts Of Rhaponticum Uniflorum(L.)DC. Flower

Rhaponticum uniflorum(L.)DC.flowers have been commonly used by Mongolian medicine in the treatment of hot heart heat,intestinal tingling,blister fever and other diseases.Based on the previous research of our research team,this paper has been carried on the research to the quality control and the anti-inflammatory effective part extraction and purification technology of the Rhaponticum uniflorum(L.)DC.flowers.The paper is divided into three parts.First of all,five flavonoid compounds were isolated and identified as luteolin,apigenin,quercetin,quercitrin and afzelin according to the anti-inflammatory properties compounds in Rhaponticum uniflorum(L.)DC.flowers.The second,based on total flavonoids,the main anti-inflammatory effective part of Rhaponticum uniflorum(L.)DC.flowers active ingredients,the UV quantification method of total flavonoids was built about Rhaponticum uniflorum(L.)DC.flowers with quercetin as reference.The linear regression equation of total flavonoids was Y= 0.064X-0.147,and r equaled to 0.9995.The linear relationship of was in the range of 4.8 μg/m L-14.4 μg/m L.Average recoveries of total flavonoids was 98.40% and RSD was 1.3%;in addition,the HPLC quantification methods of luteolin and quercitrin in Rhaponticum uniflorum(L.)DC.flowers was also built.The linear regression equation of luteolin was Y = 877.8X-788.5,and r equaled to 0.9995.The linear relationship of luteolin was in the range of 1.1 μg-22 μg.Average recoveries of luteolin was 98.71% and RSD was 1.6%;as for quercitrin,the linear regression equation was Y = 680.9X + 404,and r equaled to 0.9990.The linear relationship was in the range of 1.05 μg-21 μg.Average recoveries of quercitrin was 101.01% and RSD was 1.8%;The third,optimized extraction and purification method of Rhaponticum uniflorum(L.)DC.flowers anti-inflammatory effective part were built based on the index of total flavonoids.The best extraction technological parameter was 12 times the amount of 75% ethanol was refluxed two times,every time for 45 minutes.As for the optimal purification method,AB-8 macroporous resin was used,the concentration of the sample is 0.05 g/mL,the pH of sample was 5,the absorptive flow rate was 1 mL/min and the ratio of diameter to height was 1:9,water cleaning,and the eluant was 5 BV of 50% ethanol.The purity of total flavonoids in the anti-inflammatory effective parts of Rhaponticum uniflorum(L.)DC.flowers was 42.3% detected by UV,and the purity of luteolin and quercitrin were 3.1% and 5.8% detected by HPLC.
