Objective:To investigate the anti-tumor effect of Rhaponticum uniflorum water extract (RUWE) in H22-bearing mice research and its mechanisms.Methods:The H22 transplanted mouse models were established and randomly divided into 5 groups:model, high, medium and low dose RUWE, and 5-Fu group. Animals were treated with RUWE for 10 days, then the tumor growth inhibition ratios, spleen and thymus indexes were caluclated. The serum tumor necrosis factor-a (TNF-a) and interlekin-2 (IL-2) was determined with ELISA method, the total antioxidant capacity (T-AOC) and malondialdehyde (MDA) content were detected by colorimetric method.Use DNAladder method detection the existence of apoptosis in tumor tissue. Immunohisto-chemical method detection of tumor tissue microvessel density (MVD), and Westernblot method detection protein expression of tumor tissue vascular endothelial growth factor (VEGF),hypoxia-inducible factor-la (HIF-lα),vascular endothelial growth factor receptor2 (VEGFR2).Result 1,compare with model group, Animal experiments showed that RUWE significantly reduced xenograft tumor weight (P<0.05), Improve inhibition rate, and The RUWE high, medium and low dose inhibition rates were 38.05%,34.98%,26.95%. And spleen index increased significantly; compare with 5-Fu, Rhaponticum uniflorum can significantly enhance the mice weight before and after the experiment (P<0.05). improve Spleen index and thymux index significantly (P<0.05).2,compare with model group, Serum index test results show that RUWE can significantly increase serum IL-2and reduceTNF-a, increase serum T-AOC and reduce the MDA level.3,compare with model group, DNA agarose electrophoresis results are shown that RUWE group characteristic DNA ladder bands was obviously.4,The results of immunohistochemical showed that the levels of MVD was significantly lower in 5-Fu group than in model group(P<0.05);In middle,hight group of RUWE, In RUWE group, the levels of MVD lower significantly (P<0.05).5,compare with model group, western blot results are shown that all of VEGF,HIF-1α,VEGFR2 expression decreased in RUWE group.Conclusion:RUWE could exhibit an inhibitory effect on the growth of transplanted H22tumor, probably via the improved immune function,antioxdative capability,promoted tumor tissue apoptosis and inhibited tumor angiogenesis...
