Objective: through establishing the bone defect model of the Japanese rabbit in femoral condyle,to observe the effect of bone regeneration with the A-PRF and ?-TCP composite materials,by setting up the positive control group A-PRF,group ?-TCP and the blank control group of critical bone defect to compare.in order to figure out the features and the advantages of the A-PRF and ?-TCP composite materials in bone-repairing process,which can provide theoretical reference for clinical application.Methods: according to the different materials used in the area of bone defect,39 rabbits were randomly divided into A-PRF group,?-TCP group,A-PRF and ?-TCP composite group(12 each)and blank group(3).Making the critical bone defect with 6.0 mm in diameter and 8.0 mm in depth in femoral condyle of rabbit on hind legs,and fill in A-PRF in the group A-PRF(2000 rpm/min,centrifuging 24 min);fill in ?-TCP in the group ?-TCP;in A-PRF and ?-TCP group filled in the quality ratio of 1:1 A-PRF and ?-TCP,the blank group fill nothing,after one month,two months,three months,removing the femoral condyle bone for observation and taking X-ray pictures,then sent them for the HE staining and Masson staining and immunohistochemical detection.Results: The model of femoral condylar defect was successfully established in 39 experimental animals.In observation,over time the A-PRF group,?-TCP group,A-PRF and ?-TCP composite group have obvious bone formation,among them A-PRF and ?-TCP composite group have better influence than ?-TCP group,A-PRF group,and the blank control group is the worst.In X-ray observation,along with the extension of time for observation,A-PRF group,?-TCP group,A-PRF and ?-TCP composite group all have high density shadow,and showing the centrality trend from the range of bone defect to the center of materials,and also the high density shadow in A-PRF and ?-TCP composite group formed quickly and finished bone repairing faster compared with A-PRF group and ?-TCP group.In the blank control group,only partial high density shadow were seen at the end of the experiment.HE staining:A-PRF group,?-TCP group and A-PRF and ?-TCP composite group all formed new bone tissue,besides,at the same time point A-PRF and ?-TCP composite group had more new bone tissue than A-PRF group and ?-TCP group,the new bone tissue in blank control group was least.Masson staining: the activity of bone formation in A-PRF and ?-TCP composite group was obviously better than A-PRF group and ?-TCP group at the same time point,and also the Osteogenesis rate in A-PRF and ?-TCP composite group was higher than A-PRF group and ?-TCP group,and the blank control group was the least which indicated it has the worst ability of bone formation,by the way,?-TCP group is better than A-PRF group.the above results were statistical differences(P < 0.05).Immunohistochemical analysis: at every time point can observe that the expression of RANKL and OPG in A-PRF group,?-TCP group and A-PRF and ?-TCP composite group were better than the blank control group,the difference has statistical significance(P < 0.05),while the comparison shows that A-PRF and ?-TCP composite group is better than ?-TCP group,the difference has statistical significance(P < 0.05).Within the same group,along with the extension of time,the expression of RANKL in A-PRF group,?-TCP group and A-PRF and ?-TCP composite group was gradually increased,with statistically significant difference(P < 0.05),the expression of RANKL in blank control group increased slowly,and the expression of OPG was stable at first,and then weakened over time.Conclusion: 1.The model of the critical bone defect was successfully established in the femoral condyle.2.The results of X-ray,HE staining,Masson staining and immunohistochemical staining indicateed that A-PRF,?-TCP and the complex of the two all could promote bone tissue regeneration,and the effect of bone regeneration of the complex of A-PRF and ?-TCP was better than that of using A-PRF or ?-TCP alone at the same point.3.The regeneration in bone defect area showed centripetal trend.
