| Objective:To analyze the role and mechanism of insulin in prostate cancer cells.Methods:Selected respectively to three kinds of cells to insulin stimulation,i.e.,the PC-3,LNCaP-AI and LNCaP,insulin receptor expression change selected immunocytochemistry(ICC,immuno cyto chemistry)method,the protein immunoblot(Western Blotting)and real-time fluorescence quantitative polymerase chain reaction(Real-Time PCR)to detect;At the same time,invasive test(Transewll)selected cell,scratch test,tetrazolium salt method and cell counting method for cells to stimuli respectively before and after the attack ability,migration,vitality,and to detect the changes of proliferation;The expression of Ras,erkk1/2 and MEK1/2 protein before the stimulation was detected by Western Blotting,and the activity changes of MAPK signaling pathway were observed on this basis.Results:Insulin stimulation can increase the three types of cells in a certain extent in insulin receptor mRNA and protein expression level,also can inhibit LNCaP cell invasion,migration,vitality,and proliferation,can further promote the PC-3 cells;However,the invasion,migration,activity altered before and after the stimulation of insulin stimulation.Insulin can reduce MEK,ERK phosphorylation level and LNCaP cell Ras expression level and promote MEK,ERK phosphorylation level and pc-3 cell Ras expression level,but no significant effect on LNCaP-AI.Conclusion:With insulin receptor,insulin to Ras-MEK-ERK signaling pathway activation have an impact,and on this basis on LNCaP cell invasion,migration,vitality and proliferation inhibition effect,for the PC-3 cell invasion,migration and proliferation vitality and to promote,but the cells had no significant effect on LNCaP-AI. |
