Background: In the United States and Europe, prostate cancer (PCa) is the the mostcommon malignancy in male reproductive system, which incidence increases withage growth, ranking second in the male cancer mortality, second only to lung cancer.PCa incidence increases year by year, according to statistics, in2010in the UnitedStates,217000cases of men was diagnosed with PCa, and there are32,000maleswould die because of metastatic PCa. It is estimated that PCa the incidence willincrease2.5times by the year2050since the average life expectancy continues toincrease. In the past its incidence is low in our country, but with the emergence ofaging of population and changes in diet,its incidence is rising year by year. Theetiology of PCa is complex, the incidence exact mechanism is unclear, may berelated to lifestyle, age, race, environment, heredity, hormones and other factors. Inrecent years, more and more studies reveal that chronic inflammation of the prostatemay be an important underlying factor in the occurrence and progression of PCafrom epidemiology, pathology, molecular biology view.Objective:We want to ensure that SCF/c-kit pathway plays an import role in themigration of mast cells to prostate cancer cells,which will promote the growth ofprostate cancer cells.Methods:1.Transwell experiment will be used to observe the phenomenon of migration ofmast cells to prostate cancer cells;2MTT assay will test that what influence of prostate cancer proliferation from mast cell;3. qRT-PCR will be applied for the mRNAexpression of c-kit on mast cells and SCFon prostate cancer cells;4.Western Blot assay will be applied for the protein expression of c-kit on mast cellsand SCF on prostate cancer cells;5c-kit antibody will be used to observe the changes of the migration of mast cells toprostate cancer cells and the growth of prostate cancer cells;Results:1After co-cultured24h,we observe that prostate cancer cells can promote themigration of mast cells, the migration rate of mast cells in the experimental group (%)=10.167±0.833, and the migration rate of mast cells in the control group (%)=0.833±0.208, the difference is statistically significant (P <0.05).2MTT test:Compared with the control group, the proliferation of prostate cancer cells in theexperimental group increases with the increasing of the number of mast cells;.3qRT-PCR technology:. mRNAexpression of c-kit and SCF in the experimentalgroup was significantly higher, the difference was statistically significant (P <0.05).4.Western Blot method: protein expression of c-kit and SCF in the experimentalgroup was significantly higher, the difference was statistically significant (P <0.05).5After addition of the c-kit antibody, compared with the control group, the mastcell migration of experimental group is significantly reduced, and the proliferationof prostate cancer cells in the experimental group is significantly slower.Conclusion:1. Prostate cancer cells can promote mast cells migrate to them.2. Mast cells can promote the proliferation of prostate cancer cells after its migration.3. SCF/c-kit pathway plays an import role in the migration of mast cells to prostate cancer cells,and promote the growth of prostate cancer cells.4c-kit inhibitor can inhibit mast cells migration to prostate cancer cells, and inhibitthe growth of prostate cancer cells.
