Objective:To investigate the significance of Tissue transglutaminase in children with Henoch-Sch???nlein Purpura nephritis from two aspects,one is gene,the other one is protein.To explore the potential molecular mechanism of anaphylactoid purpura and further provide a novel theoretical foundation for clinical therapy.Methods:The author selected randomly 20 cases Henoch–Sch???nlein purpura patients and20 cases Henoch–Sch???nlein purpura nephritis patients as experimental group in Jiangxi Provincial Children's Hospital from October in 2016 to April in 2017.The all patients were for the first time in hospital.In the meantime,20 cases healthy children were included as control group with age and gender matched from child healthcare department.The mRNA expression of TGM2 in peripheral blood nucleated cells were detected in healthy controls,patients with Henoch–Sch???nlein purpura and Henoch–Sch???nlein purpura nephritis by using real-time quantitative polymerase chain reaction.Meanwhile,five cases of Wilms'tumor peripheral normal renal tissues with homogeneity in age and gender were collected from pathology department as controls.The expression of TGM2 in renal tissues was measure by immunohistochemical method in healthy controls and patients with Henoch–Sch???nlein purpura nephritis;furthermore,according to the International study of kidney disease,the Henoch–Sch???nlein purpura nephritis group were divided into different pathological grades,the author also analyzed the relationship between TGM2 and degree of renal pathological damage.Results:The mRNA level of TGM2 in peripheral blood was the lowest in control group,but highest in Henoch–Sch???nlein purpura nephritis patients.It was remarkably significant in Henoch–Sch???nlein purpura nephritis group by 5.33±1.46-fold,compared to Henoch–Sch???nlein purpura groups by 1.21±0.32-fold?P value is 0.035?and healthy control group by 1.19±0.15-fold?P value is 0.031?.However,there was no statistical significance between allergic purpura and healthy group.Accordingly,the protein level of TGM2 in renal tissue was increased in Henoch–Sch???nlein purpura nephritis to compare with controls.The inter-quartile range of immunes histochemistry positive cell score in Henoch–Sch???nlein purpura nephritis group was from 2.25 to 3.00,but was from 1.00 to 1.50 in nomal renal tissues,there is a significant difference between the two groups?P=0.001?.Moreover,according to the International study of kidney disease,Henoch–Sch???nlein purpura nephritis patients were included in 4 cases by?grade,12cases by?grade,4 cases by?grade,and the scores(P25,P75)were respectively from 2.00 to 2.75,from 3.00 to 3.00,from3.00 to 4.00;it is statistical significance among the three grades.The data revealed the expression of TGM2 in Henoch–Sch???nlein purpura nephritis with different pathological grades was different and it was positively correlated with pathological grades?P<0.001?,the Kendall correlation index was as high as 0.659.Namely,the much more serious the renal damage,the stronger the positive expression of TGM2.Conclusion:The TGM2 may be closely implicated with the occurrence of Henoch-Sch???nlein purpura kidney injury and play a crucial role in Henoch-Sch???nlein purpura nephritis.The increasing expression of TGM2 could be a indicator to evaluate that if the renal tissue was injured and the pathological grades.
