Objective:Different concentrations of procyanidins were used to culture Streptococcus mutans at different times,then detection the concentration of Streptococcus mutans?concentration of NO and the activity of NOS,and compare differences between them.The effects of grape seed procyanidins(GSPE)on Streptococcus mutans concentration?NO concentration and NOS activity in vitro were deduced,and the mechanism of grape seed procyanidins(GSPE)inhibiting Streptococcus mutans was further deduced.Method:1.Culture Streptococcus Mutans with different concentrations of Grape Seed procyanidins for 24 h.The bacterial concentration was measured at 2,3,4,5,6,8,12,14,20 and 24 hours,and the growth curve was plotted.The effect of Grape Seed procyanidins on Streptococcus mutans was clarifying.2.The NO concentration and NOS activity in the mixed solution were measured at 2,3,4,5,6,8,12,14 and 24 hours,and the change curves were drawn to determine whether the grape seed procyanidins could promote the synthesis of NO from Streptococcus mutans NOS,and the influence mode was determined.3.The relationship between bacteria concentration,NO concentration and NOS activity was analyzed in order to clarify the relationship between them.Then the experimental conjecture that the grape seed procyanidins could inhibit the growth of Streptococcus mutans by promoting NOS synthesis and NO.Result:1.Grape seed procyanidins(GSPE)could inhibit the growth of Streptococcus mutans,and the effect of GSPE on the Streptococcus mutans was statistically significant when the concentration of procyanidins(GSPE)was at 25mg/ml and 12.5mg/ml.2.Different concentrations of grape seed procyanidins(GSPE)can increase the concentration of NO and reach the peak of concentration at 4h,and gradually decrease after 4h,but it can ensure that the whole 16 h is maintained at a higher level than that of the negative control group.3.Different concentrations of grape seed procyanidins(GSPE)can increase the activity of NOS to the highest level in 2h,and can be maintained at a higher level than that in the negative control group in 4h.Conclusion:Grape seed procyanidins(GSPE)can inhibit the growth of Streptococcus mutans by promoting NOS synthesis into NO.
