| Aim:To evaluate the mRNAs and proteins expression of Interferon-induced transmembrane protein 3(IFITM3)in human HCCLM3 transfected with sh-IFITM3 and SMCC7721 transfected pcDNA3.1(+)-IFITM3.And explore the impact of IFITM3 to epithelial-mesenchymal transition in hepatocellular carcinoma(HCC).Furthermore,to detect the role of IFITM3 in the invasion and metastasis of HCC.Methods:1.Collected tumor and adjacent tissue specimens from the patients that underwent HCC resection.Using qRT-PCR,IHC and Western blot to analyze the relationship between the expression of IFITM3 gene and clinicopathological parame-ters in HCC patients2.shIFITM3 and pcDNA3.1(+)-IFITM3 were transfected into HCCLM3 and SMCC7721 cells.And western blot were used to detect the relationship between the IFITM3 gene and EMT markers.3.The cell invasion and metastasis were assessed by Transwell and Woundhealing assays,after transfection.4.Western blot were used to detect the relationship between the IFITM3 gene and p38-MAPK pathway.5.Statistical software SPSS 17.0 was used to analysis the data.Results:1.IFITM3 expression was upregulated in HCC tissues.2.IFITM3 overexpression is closely related to HCC progression.3.IFITM3 promotes HCC metastasis in vitro.4.IFITM3 regulates HCC progression by inducing EMT though p38-MAPK pathway.Conclusion:IFITM3 was upregulated in HCC tissues,and its overexpression is closely related to HCC progression.IFITM3 promotes HCC invasion and metastasis in vitro.Mechanistically,IFITM3 may exerted its function through the EMT by activating p38-MAPK signaling pathway. |
