Preliminary Research About The Role Of ITr35 Cells In The Immune Etiological Pathogenesis Of Oral Lichen Planus

Abstract/Summary:
Objective:The aim of this study was to determine the frequency of circulating Treg cells,iTr35 cells and related cytokine IL-35 levels,evaluating the pathogenesis of their possible role and significance in OLP.Methods:The peripheral blood of thirty-five patients with OLP(fifteen cases of reticular OLP and twenty cases of erosive OLP)and twenty-five healthy controls were enrolled in this study.1 Flow cytometry(FCM)was used to test CD3~+,CD4~+,CD8~+,CD19~+and CD16+56(natural killer cell,NK)subsets.Nephelometry assays were used to detect IgG,IgA,IgM,complement C3 and C4.And analysis cellular and humoral immune status in patients with OLP.2 In these OLP patients,the peripheral blood of twenty-seven patients with OLP(eleven cases of reticular OLP and sixteen cases of erosive OLP)and sixteen healthy controls were enrolled.FCM was used to detect the frequency of CD4~+Foxp3~+Treg cells and CD4~+Foxp3~-Ebi3~+p35~+iTr35 cells.And analysis the relationship between the two kinds of cells.3 IL-35 contents in serum of patients with OLP and normal controls were detected by enzyme linked immunosorbent assay(ELISA)and analyze its relationship with immunity.Results:1 The results show that in OLP group,the subsets of CD8,NK cells and complement C3,C4 were lower than normal value.However,the subsets of CD19cells and IgG,IgM were higher than normal value.2(1)The results revealed a higher expressions of CD4~+Foxp3~+Treg in erosive OLP 3.503(2.355~4.200)%and reticular OLP 2.850(2.480~3.070)%when compared with normal controls 1.955(1.733~2.123)%,but there was no significance between erosive OLP and reticular OLP.(2)The results showed significantly elevated CD4~+Foxp3~-Ebi3~+p35~+iTr35 cells in erosive OLP0.124(0.046~0.185)%and reticular OLP 0.058(0.031~0.078)%when compared with normal controls 0.029(0.016~0.040)%.However,there was no significance between reticular OLP and erosive OLP.(3)There were positive correlations exsist between iTr35cells and CD4~+Foxp3~+Treg cells(r=0.385,P=0.048),iTr35 cells and IL-35 producing cells in CD4~+Foxp3~+Treg cells(r=0.723,P<0.001).3(1)The expressions of serum IL-35levels both in erosive OLP(38.956±8.945)and reticular OLP(37.454±8.124)were higher than those in normal controls(32.069±4.11),but there there was no significance between reticular OLP and erosive OLP.(2)Pearson correlation analysis showed that there were positive correlations between serum IL-35 levels and IgG(r=0.583,P<0.001),whereas no significant linear correlations between serum IL-35 levels and iTr35 cells.Conclusion:1 Low cellular immunity accompanied by a certain degree of humoral immune disorder exists in patients with OLP.2 Increased frequency of CD4~+Foxp3~-Ebi3~+p35~+iTr35 cells may be an important pathogenesis of OLP,in which CD4~+Foxp3~+Tregs may secrete IL-35to promote iTr35 formation.3 The high expression of inhibitory cytokine IL-35 may be involved in the immunomodulation of OLP by regulating mechanism through negative feedback.
