Objective(1)To asses safety of Shaqi capsule by acute toxicity test.(2)To study the hypoglycemic effect and mechanism of Shaqi capsule by a model rat of insulin resistance.Methods(1)(1)20 kunming mice weighing 18-22 g were fasted 12 h before drug administration.Shaqi capsule was given orally at 20 m L/kg via gastric gavage to mice,and nothing unusual was found.Then,the maximum tolerated dose of shaqi capsule was determined to be 60g/kg.(2)Wistar rats were fed with enrich-diet for 32 days,then STZ was injected abdominally.After that the rats were treated with S(supernatant),R(residue)and SC(Shaqi capsule)for 4 weeks respectively,then blood biochemical indexes(insulin,C-peptide,Hb Alc?GSP,CHO?TG?HDL-C?LDL-C and pancreas were tested to observe the effect of Shaqi capsule.Results(1)Shaqi capsule was given orally at 20 m L/kg via gastric gavage to mice,equivalent to 126 times of a clinical human dose.and nothing unusual was found.Then,(2)S,R and SC all could reduce the level of blood glucose in varying degrees.The levels of insulin and C-peptide in blank group and model group were not significantly different.Shaqi capsule could significantly reduce the levels of TG,and MDA,and increase the level of SOD.Conclusion(1)Shaqi capsule did not produce acute toxicity in mice.(2)S,R and SC all could reduce the level of blood glucose in varying degrees.
