In this paper,Cress was used as raw material to prepare 5% water cress ultrafine powder feed and water extract respectively.The high-dose streptozotocin(STZ)was used to induce type 2 diabetic mice as a model to explore the effect of Cress improve insulin resistance and anti-oxidative stress.The specific research results are as follows:1.After using the ultrafine powder feed and water extract of water cress for four weeks,the effects on body weight(BW),fasting blood glucose level(FBG),blood lipid level,organ index and pancreatic pathological section were investigated.The results showed that after 4 weeks of cress intervention,the blood glucose of diabetic mice decreased,the weight loss was alleviated,and the levels of total cholesterol(TC),triglyceride(TG)and low density lipoprotein(LDL-C)in the serum of diabetic mice.Significantly decreased,while high-density lipoprotein(HDL-C)levels were significantly increased,while cress treatment was effective to reduce the organ index of diabetic mice and repair pancreatic tissue.Therefore,cress has the effect of lowering blood sugar,regulating blood lipids,relieving weight loss,and repairing pancreatic tissue damage in diabetic mice.2.To study the degree of antioxidative activity enzymes,oxidation products and DNA fragmentation in liver tissues of diabetic mice,and to explore the effect of Cress on anti-oxidative stress in diabetes.The results showed that the levels of superoxide dismutase(SOD),catalase(CAT),glutathione(GSH)and glutathione catalase(GSH-PX)in diabetic mice were significant.Increased;the content of malondialdehyde(MDA)and lipid peroxide(LPO)decreased significantly,and the degree of DNA fragmentation in liver cells was significantly reduced.Therefore,Cress can effectively improve the antioxidant activity in diabetic mice and repair DNA damage.The effects of Cress on insulin resistance in diabetic mice were studied.The insulin resistance index was determined and the insulin resistance was determined.The related genes PTEN,IRS-1,IRS-2 were determined by semi-quantitative RT-PCR.The expression of PI3 K,Akt-2 and GLUT4 was simultaneously determined by IRS-1,PI3 K and Akt-2 genes.The results showed that the serum insulin content and insulin resistance index of diabetic mice decreased significantly after four weeks of cress treatment.The mRNA expression levels of IRS-2,PI3 K,Akt and GLUT4 were significantly increased in the watercress treatment group,among which IRS-1,the protein expression levels of PI3 K and GLUT4 were significantly increased.Therefore,cress may enhance the absorption and utilization of glucose in diabetic mice by enhancing the gene expression of key factors IRS-1,IRS-2,PI3 K,Akt-2 and GLUT4 in the PI3K/Akt signaling pathway,thereby improving diabetic mice.Insulin resistance symptoms.
