Overexpression Of MiR-221/miR-222 In The Cerebrospinal Fluid Of Glioma Patients And The Potential Diagnostic Value

Objective:To investigate the expression of miR-221/miR-222 in the cerebrospinal fluid(CSF),and the differences of mi R-221/mi R-222 expression between glioma patients and non-glioma controls were compared to further explore the potential value of miR-221 and miR-222 in CSF for diagnosing glioma.Methods:A total of 64 cases of cerebrospinal fluid samples were enrolled in our study,consist of 31 glioma patients and 33 non-glioma patients(controls)from the neurosurgery apartment of the second affiliated hospital of Nanjing medical university during January 2013 to February 2017.The case group was subdivided into 17 cases of high grade group and 14 cases of low grade group.Quantitative reverse transcription polymerase chain reaction(qRT-PCR)were used to detecting the expression of mi R-221 and miR-222 in each CSF sample.Then the expression levels of each group were comparaed to statistically analysis the differences by the SPSS software.Further more,diagnostic value of miR-221 and miR-222 in CSF were assessed by receiver operating characteristic(ROC)curve.Results:The expression of miR-221 was 2.89-fold higher in CSF of glioma group comparing with control group(P<0.001)and the expression of miR-222 was 2.38-fold higher than control group(P<0.001).The miR-221 expression in the high grade group was 1.37-fold higher than in low grade group(P=0.186,no statistics significance),while the difference of miR-222 was statistically significant(1.49-fold,P=0.007).The area under the ROC curve(AUC)of miR-221 was 0.831,and the AUC of mi R-222 was 0.773,which means both of them can separate the case group from the control group.The AUC of the combination of miR-221/mi R-222 family was 0.849,better than single miR-221 or mi R-222 in diagnostic efficacy.Conclusion:The expression level of miR-221/miR-222 in CSF of glioma patients was significantly higher than that of control group,and miR-222 expression in high grade glioma CSF was obviously higher than that of low grade group.The study indicated that miR-221/miR-222 in CSF could serve as a potential biomarker in diagnosing glioma and it worth more samples to further analysis,as well as horizontal comparison of multiple specimens source to verify the value.
