ObjectiveTo detect the expression levels of?3GalT2 protein in breast cancer,and assess its relationship with clinicopathological features,and understand the role of?3GalT2 gene in breast cancer cell apoptosis.MethodsThe expression of?3GalT2 protein in breast cancer tissues was detected by SP immunohistochemistry and Western blot,analyzing the relationship between the expression of?3GalT2 protein and clinicopathological factors in breast cancer patients.The expression of?3GalT2 mRNA in breast cancer was analyzed in Oncomine database,the relationship between the?3GalT2expression levels and prognosis in patients with breast cancer was studied by Kaplan Meier method.The effect of up-regulated?3GalT2 gene on the biological function of breast cancer cell line MCF-7 was detected by Western blot.The colocalization of?3GalT2 protein with mitochondria was observed by immunofluorescence confocal microscopy.ResultsImmunohistochemistry and Western blot detection results show that the expression of?3GalT2 protein in breast cancer was lower than that in normal breast tissues(48.00%,24/50).The positive rate of?3GalT2 protein in cancer tissue was 82.00%(42/50),the difference was statistically significant(?~2=100.00,P=0.000);?3GalT2 protein was correlated with clinicopathological factors(lymph node metastasis,tumor differentiation and staging)(P<0.05);high expression of?3GalT2 protein promote the apoptosis of MCF-7 cells occurs;the?3GalT2 protein is located on the mitochondria of breast cancer cells.ConclusionsThe decreased expression of?3GalT2 protein in breast cancer tissues suggests that?3GalT2 may be involved in the formation of breast tumors;Overexpression of?3GalT2 protein promotes apoptosis of breast tumor cells,suggesting that this gene may inhibit the development of breast cancer;the expression of?3GalT2 is associated with clinical factors(metastasis of lymph nodes,tumor differentiation,staging)in Brc patients,suggesting that it may be involved in malignant biological behaviors of breast cancer and may be a molecular marker for judging the prognosis of Brc;?3GalT2 protein is localized on the mitochondria in the cytoplasm and overexpressed.This gene activates the mitochondrial apoptotic pathway,leading to apoptosis of breast tumor cells.
