Objective To investigate the effecs of the Augmented Xiaoyao Powder in treating slow transit constipation rats and Cholagogic effect.Methods 48 SD rats were randomly divided into 6 groups,including normal control group,model control group,Augmented Xiaoyao Powder high-dose group,Augmented Xiaoyao Powder middle-dose group,Augmented Xiaoyao Powder low-dose group and Ursodeoxycholic Acid group.The slow transit constipation rat models were established by using compound diphenoxylate for 14 days without interruption.And then drug treatment was given after the successful establishment of the model.The dose of the drug was calculated according to the conversion coefficient between human and rat.Normal control group and model control group were given physiological saline,Augmented Xiaoyao Powder high-dose group(45.5g/kg),Augmented Xiaoyao Powder middle-dose group(22.75g/kg),Augmented Xiaoyao Powder low-dose group(11.375g/kg)and Ursodeoxycholic Acid group(62.5mg/kg).At the end of the treatment,the intestinal transport function and the dry wet mass ratio of feces were measured in rats of each group.And then Bile drainage by bile duct intubation,bile drainage.Finially the bile drainage flow was measured,and the total bile acid concentration in feces,serum and bile samples was determined by enzyme-labeling instrument.Results 1.Compared with the normal control group,there was no significant difference in before modeling,after modeling and after treatment of the each group’s rats weight(P>0.05).2.Compared with the model control group,the time of gastrointestinal transmission and the ratio of the Fecal dry wet mass were significantly reduced in the Augmented Xiaoyao Powder middle-dose group(P<0.01).3.Compared with the model control group,the volume of bile secretion increased in the normal control group and the Augmented Xiaoyao Powder middle-dose group at 1h,2h,3h and 4h respectively(P<0.05,P<0.01).4.Compared with the model control group of the male rats,the concentration of TBA in bile was increased in the normal control group,the Augmented Xiaoyao Powder low-dose group and the Augmented Xiaoyao Powder middle-dose group(P<0.05),the concentration of TBA in serum was increased in the Augmented Xiaoyao Powder high-dose group(P<0.05),and there was no significant difference in the concentration of TBA in feces(P>0.05).Compared with the model control group of the female rats,There was no significant difference in the concentration of TBA in serum and feces.Conclusion The Augmented Xiaoyao Powder middle-dose group can shorten the time of gastrointestinal transit,reduce the dry and wet mass ratio of feces,increase the bile secretion and regulate the bile composition in male rats.The Augmented Xiaoyao Powder middle-dose group can promote intestinal peristalsis and intestinal secretion through cholagogic effect,and plays a role in treating slow transit constipation.The Xiaoyao Powder can increase the concentration of TB A in the serum of male rats,and it is related to the concentration of drug,whether it is related to the damage of hepatocytes,which needs further toxicological study to verify.Compared with the control group,there was no significant difference in the concentration of TBA in feces,suggesting that the concentration of bile acid in colon might not be increased.
