Objective:To investigate the relationship between HPV infection and EGFR,and between P16 and EGFR.To explore the difference of P16 and EGFR protein expression and the clinicopathological features as well as the infection of HPV in oral and oropharyngeal squamous cell carcinoma.Method:Tissues sample of 53 oral squamous cell carcinoma and 35 oro pharyngeal squamous cell carcinoma were collected.Immunohistochemistry was used to detect the expression of P16 and EGFR protein in oral and oropharyngeal squamous cell carcinoma.In Situ Hybridization was used for the detection of HPV infection.The data were analyzed using the ttest,the?~2 test and the relevance analysis of categorical variables.Results:The positive rate of P16 was 40.9%,and the positive rate of EGFR was 69.3%.P16 and EGFR were correlated in oral and oropharyngeal squamous cell carcinoma(P<0.05,?~2=14.77,r=0.379).The positive rate of P16 was 22.6%(12/53)in oral squamous cell carcinoma and 68.6%(24/35)in patients with oropharyngeal squamous cell carcinoma,and the difference was statistically significant(P<0.000).The positive rate of EGFR was 56.6%(30/53)in oralsquamous cell carcinoma,but was 88.6%(31/35)in oropharyngeal squamous cell carcinoma,and the difference was statistically significant(P<0.001).P16 positive group of patients with smoking,tumor differentiation,recurrence and gender compares with negative group,there was statistically difference(P<0.05).The difference between EGFR positive group and negative group on gender was statistically significant(P<0.05).The total infection rate of HPV in oral and oropharyngeal squamous cell carcinoma was 18.2%(16/88),of which 15.1%(8/53)in oral squamous cell carcinoma and 22.9%(8/35)in oropharyngeal squamous cell carcinoma.HPV infection was not related to age,smoking,alcohol consumption,tumor site,tumor differentiation,recurrence,lymph node metastasis,T stage,N stage,and clinical staging(P>0.05).But it had tremendous statistically significance with sex,P16 protein expression and EGFR expression(P<0.05).There were 12 cases of P16 positive and HPV infection,around 33.3%(12/36).HPV infection was associated with EGFR expression(P<0.05,?~22=5.488,r=0.258).Conclusion:In oral and oropharyngeal squamous cell carcinoma,there exists a positive link between HPV infection and EGFR,as well as between P16 and EGFR.The expression of P16 and EGFR were different in oral and oropharyngeal squamous cell carcinoma.In oral and oropharyngeal squamous cell carcinoma,the distribution of HPV infection is different,and the infection rate of HPV in oropharynx is greater than that of the oral cavity.P16 expression was associated with tumor recurrence,differen tiation,patient sex and smoking.HPV infection was relevant to sex,P16protein expression and EGFR expression.
