Objective: Poly(C)-binding protein 1(PCBP1)is a member of the polycytosine-binding protein family and binds to poly-cytosine-rich nucleotide sequences,involved in transcriptional regulation,mRNA stability,EMT,tumor progression and other processes.Some studies have found that PCBP1 is involved in the regulation of the selective splicing process of cancer stem cell marker CD44,promoting the occurrence,development,and metastasis of tumors.However,the relationship between PCBP1 and CD44 in early non-small cell lung cancer is rarely reported.The purpose of this study was to:1.Examine the expression of PCBP1 and CD44 in 75 patients confirmed the diagnosis of stage I and II NSCLC according to pathological reports.And analyze the relationship between the clinicopathological features and proteins' expression.2.Analyze the relationship between the expression of PCBP1 and CD44 protein and the prognosis of patients with early non-small cell lung cancer.OBJECTIVE: Poly(C)-binding protein NSCLC patients were collected.The EnVision method of immunohistochemistry was used to study the expression of PCBP1 and CD44 in lung cancer tissues.SPSS22.0 software was used for statistical analysis.Survival analysis was performed using kaplan-meier method.Survival analysis was performed using the Kaplan-Meier method.The univariate analysis was performed using the log-rank test to compare the significance levels.Variables that were significant for univariate analysis were introduced into the COX multivariate through the method of gradual introduction of variables.Regression model to determine the independent factors that affect the prognosis.Result:(1)The positive expression rates of PCBP1 and CD44 in early stage NSCLC tissues were 45.61% and 20%,respectively;(2)The expression of PCBP1 protein was correlated with pathological grade and TNM stage(p<0.05);CD44 protein expression was related withpathological grade and TNM stage too(p<0.05);both were not related to gender,age,smoking history,tumor location and type,tumor size,lymph node metastasis(P>0.05);(3)Spearman rank correlation test results showed that PCBP1 had a significant negative correlation with CD44 protein expression(r=-0.508,P<0.001);(4)The Kaplan-Meier method was used to plot the survival curve.Compared with the PCBP1 positive expression group,the PCBP1 negative expression group had a lower overall survival curve(?2 =4.305 P=0.038);The CD44 positive expression group had a lower overall survival curve compared with the CD44 negative expression group(?2=4.374 P=0.036);(5)Univariate analysis showed survival time in patients with early stage NSCLC,was significantly correlated PCBP1,CD44,tumor size and TNM stage;progressive Cox multivariate regression analysis showed that survival time was significantly associated with tumor size(RR = 2.548),CD44 protein expression(RR = 3.206),and they were independent risk factors of the prognosis of early stage NSCLC patients.Conclusion: The positive expression rates of PCBP1 and CD44 in early nonsmall cell lung cancer were 45.61% and 20%,respectively.The expression of both were closely correlated with pathological grade and TNM stage.The positive expression rate of PCBP1 was high in patients with high degree of tumor differentiation and stage I.,CD44 expression rate is low,suggesting that PCBP1 may be involved in the regulation of the innitiation and development of early stage non-small cell lung cancer;Cox regression analysis shows that CD44 and tumor size are independent risk factors affecting the prognosis of early stage non-small cell lung cancer.The expression of CD44 and PCBP1 was correlated strongly.This correlation is consistent with PCBP1 involved in regulation of the selective splicing process of CD44,a tumor stem cell marker;and the prognosis of lung cancer patients with high PCBP1 expression and negative CD44 expression is better than those with low expression of PCBP1 and high expression of CD44,suggesting that PCBP1 can be used as early judgments,indicator of prognosis of lung cancer.
