ObjectivesThis research building a model of insulin resistance in obese rats,induced by high fat diet.To observe expression of the SIRT1/PPAR? in white adipose tissue of insulin resistance in obese rats via electricacupuncture method,and to investigate the related mechanism of electricacupuncture to improve the insulin resistance in obesity.MethodsSixty male wistar rats at the age of eight weeks,were randomly divided into normal control group(n=16)and high fat diet group(n=48)after feeding adaptively for a week.The normal control group were given ordinary diet,and the high fat diet group were given high fat diet for 8 weeks.Then 6 rats in normal control group and 24 rats in high fat diet group were randomly selected to participate in high insulin regularglucose clamps,which was used to assess the insulin resistance,meanwhile,calculating the Lee 's index of two groups to evaluate the obesity.After the models were established,the high fat diet group were randomly divided into model group,electricacupuncture group,sham electricacupuncture group,resveratrol group,each group of 6.Giving ordinary diet in addition to the ordinary group,other groups were given high fat diet.Normal control group and model group were given nothing therapy,electricacupuncture group were given electricacupuncture therapy 3 times a week for 8 weeks,sham electricacupuncture group were given acupuncture therapy without electricity 3 times a week for 8 weeks,resveratrol group were given by gavage 5 times a week for 8 weeks.After intervention with eight weeks,weight and body length of each rat were measured and Lee'S index were figured out,fasting blood-glucose and postprandial blood sugar of each rat were measured,three rats in each group were randomly selected to participate in high insulin-regularglucose clamps to calculate GIR,TC?TG?FFA were measured via Elisa kit,what's more,the expression of protein and mRNA of SIRT1,PPAR?and PRDM16 were measured by western blot and RT-PCR.Finally,the data were analysed statistically.Results1.Effects on rat weight,Lee'S index and GIR after high fat feeding for 8 weeksThe weight and Lee'S index in high fat feeding group were significantly higher than normal group(P<0.01),and GIR in high fat feeding group were significantly lower than normal group(P<0.01)after feeding of eight weeks.2.Effects on rat weight,Lee ' S index and GIR after electricacupuncture intervention for 8 weeksCompared with model group,the weight of rats of electricacupuncture group,sham electricacupuncture group and resveratrol group were significantly decreased(P<0.01),compared with sham electricacupuncture group,the weight of rats,Lee'S index and GIR of electricacupuncture group and resveratrol group were significantly decreased(P<0.01),and electricacupuncture group and resveratrol group has no statistical significance(P>0.05).3.Effects of electricacupuncture on FBG,PBG,the level of FFA,TC,TG in insulin resistance in obese ratsBefore the treatment,In comparison to normal control group,the FBG and PBG of model group,electricacupuncture group,sham electricacupuncture group and resveratrol group were significantly increased(P<0.01),at the same time,other group have no statistical significance other than the normal control group.After the treatment,In comparison to normal control group,the FBG and PBG of electricacupuncture group and resveratrol group were significantly declined(P<0.01).Compared with model group,the PBG of sham electricacupuncture group were significantlydeclined(P<0.01),while the FBG in model group and sham electricacupuncture group have no statistical significance(P>0.05).Electricacupuncture group and resveratrol group have no statistical significance(P>0.05).In comparison to model group,the level of FFA,TC and TG in electricacupuncture group and resveratrol group were significantly declined(P<0.01).electricacupuncture group and resveratrol group have no statistical significance(P>0.05),and sham electricacupuncture group and model group have no statistical significance(P>0.05).4.Effects of electricacupuncture on the level of protein and m RNA expression of SIRT1,PPAR ?,PRDM16 in insulin resistance in obese ratsIn comparison to normal control group,the level of protein and mRNA expression of SIRT1 in electricacupuncture group,sham electricacupuncture group and resveratrol group were obviously decreased(P<0.01).In comparison to model group,electricacupuncture group and resveratrol group were obviously increased(P<0.01).In comparison to resveratrol group,electricacupuncture group were obviously increased(P<0.01).Sham electricacupuncture group and model group have no statistical significance(P>0.05).In comparison to normal control group,the level of protein and mRNA expression of PPAR ? in electricacupuncture group,sham electricacupuncture group and resveratrol group were obviously decreased(P<0.05).In comparison to model group,electricacupuncture group and resveratrol group were obviously increased(P<0.01).What'more,the level of protein and mRNA expression of PPAR? in sham electricacupuncture group and model group,they have no statistical significance(P>0.05).In comparison to normal control group,the level of protein and mRNA expression of PRDM16 in electricacupuncture group,sham electricacupuncture group and resveratrol group were significantly increased(P<0.05).In comparison to model group,electricacupuncture group and resveratrol group were obviously increased(P<0.05).Sham electricacupuncture group and model group have no statistical significance(P>0.05).Conclusions1.The body weight,lee's index,the level of TC,TG,FFA,FBG and PBG can be reduced by electricacupuncture,meanwhile,insulin resistance of the insulin resistance in obese rats can be improved.2.The expression of protein and mRNA of SIRT1 in insulin resistance in obese rats can be promoted by electricacupuncture,meanwhile,that of PPAR? and PRDM16 can be promoted,and then electricacupuncture promote adipocyte differentiation and regulate energy metabolism,which may be the mechanism of improving insulin resistance in obese.
