ObjectiveThis article analyzes and characterizes the anti-oxidant active ingredients related to the treatment of diabetes in the silkworm.It provides a chemical basis for the clinical application of silkworm in the treatment of diabetes and provides a new method for the basic research of hypoglycemic substances.MethodsUsing the principle of anti-oxidation component to make KMnO4 fade,the content of anti-oxidation active ingredients in the serum of Bombyx mori,silkworm different extracts and silkworms was determined by spectrophotometric method;then,chromatographic and color-mass spectrometry techniques such as TLC and HPLC were used.In combination with the analysis of antioxidant activity,further studies were conducted on the effective sites and active ingredients of silkworms in the treatment of diabetes mellitus to analyze and confirm the anti-oxidant efficacy substance basis of silkworms for the diagnosis and treatment of diabetes.Finally,the quality control of silkworm was discussed.ResultsThe content of antioxidant active ingredients in silkworm,Bombyx mori,was determined as follows:Fujian>Jiangxi>Zhejiang;the contents of antioxidants in silkworm samples of different origins were in the same order:70%ethanol fraction>N-butanol site>ethyl acetate site>petroleum ether site.The content of antioxidants in silkworm increased gradually with the increase of the time of blood infiltration.The antioxidant active ingredients in the silkworm react with KMnO4 and appear faded spots on the silicone G plate.Based on this comparison,qualitative chromatographic peaks of metabolites in silkworm were found by comparison of HPLC profiles and qualitative analysis ofLC-MS.The results of GC-MS characterization of hypoglycemic components in silkworm showed that there were 288 compounds in 70%of silkworm and 64 compounds were identified.There were 277 compounds of total ethanol extract from silkworm,and 61 compounds were identified.TLC,UV and other methods were used to initially establish relevant quality control methods for silkworms.ConclusionsThrough this experiment,it was confirmed that the silkworm has potential antioxidant active ingredients.In this process,the thin-layer chromatography was used to analyze the antioxidant constituents,and the potential antioxidant components were analyzed and identified by HPLC,LC/MS-QTOF techniques,and related quality control methods for silkworms were initially established..The results of this paper lay a foundation for the application and development of silkworm,and provide scientific basis for the clinical application of silkworm in the treatment of diabetes.
