Objective Todisscus the preventive treatment of multiple level of Jia-Jian-Yi-Jing-Decoction on learning and memory in ovariectomized rats and its mechanism by used behavioral science,biopsy and biochemical methods,thus certain theoretical basis of its clinically used to prevent and treatment of alzheimer's disease caused by after menopause.Methods To build the rat model with alzheimer's disease by the method of bilateral ovaries castration.60 SD rats(220 ± 30)g were randomly divided into control group,model group,low dose group,middle dose group,high dose group and Positive control group.Each group of 10.After a week of build the rat model successfully,Chinese traditional medicine group was intragastric administration by using the corresponding dose of liquid medicine,estrogen group was intragastric administration by pentanoic acid estradiol,model and control group was intragastric administration by warm water.Each group was intragastric administration twice time every day for 8 weeks.Water maze training implemented in the last week,after that remove the brain tissue in rats to store in the 4% paraformaldehyde,fresh hippocampus stored at 80? below zero in the fridge.Using immunohistochemistry and Western blot,detect the expression on hippocampus in ovariectomized rats Tau,GSK-3? and ER?;using Golgi staining and Nissl staining to observe its effect on protection of neural development.Results Jia-Jian-Yi-Jing-Decoction can shorten the time of castration rats found platform and increase rats' movement distance and time in the target quadrant.It also can effectively reduce the hippocampus of Tau rats of phosphorylation and reduce the GSK-3? activity,increase levels of the ER?,improve the number and density of the axon spines,increase the density and quantity of new neurons in the hippocampus.Conclusions Jia-Jian-Yi-Jing-Decoction can effectively improve the learning and memory impairment in ovariectomized rats,protect neural development,its mechanism may be through inhibition of castration induced by GSK-3? high activity to inhibit phosphorylation of Tau protein,play a role of a kind of estrogen,and then improve the structure and function of hippocampal neurons and increase the density of dendritic spines,increase the ability of learning and memory the space.
