Objective: Study the treatment of multiple level of "Chang Yu Er Xian Decoction on learning and memory in ovariectomized rats and its mechanism.Methods: These SD rats of 60 female were selected,all weighing up to 200 grams.50 of them were castrated,and the other were sham operated(n=10).50 rats were randomly divided into model group,low dose group,middle dose group,high dose group and Positive control group.Low dose group,middle dose group,high dose group respectively given "Chang Yu Er Xian Decoction" by gavage,sham operation group and model group were given equal volume of distilled water,the positive control group with was given valerate suspension gavage.Share the medicine for 6 weeks,sixth weeks and water maze training for 5 days,a day after the interval,Morris water maze test in rats in the drug medication on the last day of the spatial learning and memory ability test.At the end of the test,respectively fetch hippocampus tissue,uterine tissue tissue,4% paraformaldehyde perfusion and Golgi staining.The preserved tissue samples,using immunohistochemistry and Western blot,detect the expression and analysis of "Chang Yu Er Xian Decoction" excessive phosphate on hippocampus in ovariectomized rats Tau and GSK-3β;using Golgi staining and Nissl staining to observe its effect on protection of neural development.Results: 1 behavioral tests showed that "Chang Yu Er Xian Decoction" can increase the water maze of rats in target quadrant motion time and distance,shorten the time for the target platform;2 Western blot showed that it can effectively reduce the hippocampus of Tau rats in p S396,p S404,p T231 and other sites of phosphorylation,and reduce the GSK-3β activity.3 immunohistochemistry showed that different doses could significantly reduce the level of Tau phosphorylation in the hippocampus of p S404 rats.4 Golgi staining showed that the dendritic spines in the hippocampus of the traditional Chinese medicine group were longer and more branched.5 Nissl staining showed that the number of Chinese medicine group hippocampal neuron Nissl’s body increased,cell morphology significantly improved.All the above results were analyzed by SPSS(p<0.05).Conclusion: "Chang Yu Er Xian Decoction" can effectively improve the learning and memory impairment in ovariectomized rats,protect neural development,its mechanism may be through inhibition of castration induced by GSK-3β high activity to inhibit phosphorylation of Tau protein,and then improve the structure and function of hippocampal neurons and increase the density of dendritic spines,increase the ability of learning and memory the space.
