Objective:To establish a mouse model of gallbladder cholesterol stone.Then use the chaihu shugan san group,Longdan Xiegan decoction,chaihu shugan san group and Longdan Xiegan Decoction orally,and the changes of serum total cholesterol and triglycerides in the observation of stone formation,content and expression of cholecystokinin,gallbladder tissue calmodulin,three phosphoinositide receptor quantity.To explore the effect of liver soothing,clearing and humid heat in the treatment of CS mice and the effect on gallbladder motility.Methods:36 C57BL mice were divided into 6 blank control group,30 model group,the normal control group was fed with full grain feed,the model group was fed with high-fat stone feed to establish cholesterol gallstone model,and 12 weeks later,the 6 mice in the model group were selected to test whether the model was successful.After successful modeling,the model mice were randomly divided into model group(MG),chaihu shugan san group(CHSG)and longdan xiegan tang group(LDXG),combined treatment group(CTG),6 rats in each group,continuous dose 3 weeks(blank control group and model control group were given saline,drug intervention group were given chaihu shugan san.longdan xiegan tang,chaihu shugan san and longdan xiegan tang).The formation rate of stone in each group was compared.Serum TC and TG contents were detected by single reagent colorimetric method.Serum CCK level was detected by ELISA method.Pathological changes in gallbladder tissues were observed by HE staining,and CaM and IP3R expression levels were detected by immunohistochemistry.Results:The blank control group,MG,CHSG and LDXG,CTG stone rates were 0%.100%.66.67%,50%,50%.Compared with the MG,LDXG and CTG TC,TG decreased significantly(P<0.05),CHSG TG significantly decreased(P<0.05);thickening of gallbladder mucosa under light microscope.MG mice,mucosa,lamina propria connective tissue,blood,tubular collagen fiber the expansion increased,and the drug intervention group were recovered.Compared with the MG,the three groups of drug intervention group significantly increased the level of CCK(P<0.05),the highest level in the CTG was statistically significant compared with the CHSG(P<0.05).Compared with the MG,the three groups of drug intervention group could significantly increase the expression level of CaM and IP3R(P<0.05).The expression level of CaM and IP3R in CTG was higher than that in CHSG,and the difference was statistically significant(P<0.05).Conclusion:Soothing the liver,eiminating dampness and heat can improve the cholesterol gallstone in mice by increasing the contraction of the gallbladder.There are two mechanisms of action,one is to reduce the concentration of TC and TG in serum lipids,the other is to increase serum CCK and IP3R and CaM levels in gallbladder tissue.Finally,it can improve the function of gallbladder contraction and exert the function of gallbladder removing stone.It is suggested that stagnation of liver qi and qi stagnation,liver and gallbladder damp heat are the important pathological bases for the formation of CS.
