Objective The establishment of 10 batches of different batch of the New yixinningshen tablets fingerprints,the effect of the sedative hypnotic effect on the rats model of insomnia were evaluated.Preliminary study on the sedative and hypnotic mechanism of New yixinningshen tablets.Combing the fingerprints with phamacodynamics to expound the relationship between fingerprint of New yixinningshen tablets and sedative and hypnotic effects and the spectral efficiency is analyzed.Methods Study on fingerprint of New yixinningshen tablets: High performance liquid chromatography(HPLC)was used to analyze 10 batches of different batch of New yixinningshen tablets.Screening for the detection wavelength,the composition of the mobile phase and the gradient elution conditions respectively and completed the methodological review.The establishment of 10 batches of different batch of the New yixinningshen tablets fingerprints,the application of the National Phermacopoeia Commission promulgated the “Chinese medicine chromatographic fingerprint similarity evaluation system software(2004A version)” 10 batches of different batch of the New yixinningshen tablets fingerprints were evaluated by similarity.New yixinningshen tablets' Pharmacodynamic evaluation of the sedative and hypnotic effects: Establishment of the model of insomnia rats by Intraperitioneal injection of PCPA.To observe the general state,body weight,sleep latency and sleep time of rats in each group under the action of pentobarbital sodium in the model group.New hypnotic effects of New yixinningshen tablets: ELISA method for the determination of 5-HT in rat hippocampus Using the grey correlation analysis method,the relative peak areas of the peaks of the combination of 10 batches of New yixinningshen tablets for different batches In the value and pharmacodynamic experiments,the increase in 5-HT content,the composition represented by each common chemical substance peaks under sedative and hypnotic effects.Results In this paper,the determination conditions of 10 batches of New yixinningshen tablets fingerprint were confirmed,10 batches of New yixinningshen tablets fingerprint were established,and 25 common peaks were obtained.Comparing with the fingerprint spectrum of reference substance,this study identified the peak 8,11,12,14,18 and 21 respectively,which is the rutin,quercetin,quercitrin,Ganodericacid A,schisandrol A and schisandrol B sequentially.This detection method has great precision,stability and repeatability.Pharmacodynamics experiment found that intraperitoneal injection PCPA method can successfully established the insomnia rats model by pentobarbital sodium synergistic experiment.Comparing with the blank group,the sleep latency of model group rats was significantly lengthened,the sleep time of model group rats was significantly shortened and the high,medium and low dose group of the New yixinningshen tablets can obviously shorten the sleep latency of the insomnia rats,and prolonged the sleep time of insomnia rats.The mechanism research found that,through HE staining and observing the structural changes of nerve cells,immunohistochemical results showed that the expression levels of 5-HT and GABA in the lower,middle and high-dose group of the New yixinningshen tablets were significantly increased,compared with those in the model group,and the lower,middle and high-dose group of the New yixinningshen tablets significantly increased the contents of 5-HT and GABA in the brains of insomnia rats,and decreased the contents of DA and NE in insomnia rats.Based on the study of the correlation between 25 common characteristic peaks and sedative hypnotic effects in 10 batches of New yixinningshen tablets fingerprints,fingerprints represent the contribution of the sequence of hypnotic and hypnotic effects and are a common feature of peak chemical component is:is: X11>X18>X21>X12>X14>X6>X19>X24>X7>X10>X9>X8>X20>X23>X25>X5>X16>X17>X22>X2>X1>X15>X4>X3>X13.Conclusions In this study,the fingerprints of New yixiningshen tablets extracted from 12 batches of different habitats were established,and combined with sedative and hypnotic effect.By using grey correlation analysis method,this study identified the size of the common characteristic peak of the New yixiningshen tablets to the sedative and hypnotic effects,and laid the foundation for further exploring the effective material of the sedative and hypnotic effect of the New yixiningshen tablets.Through the study of spectrum-effect relationship,it can reflect the characteristics of TCM compound integrity,and provide new ideas and experimental basis for the research on the spectrum-activity relationship of chinese herbal medicine compound preparation.
