ObjectiveTo detect the expressions of CD63 and Fc?RI in blood eosinophils from allergic rhinitis(AR)patients,chronic simple rhinitis patients and healthy controls(HCs)in rest state or after allergen stimulation.Finally,we evaluated the meaning of eosinophils activation test in diagnosis of allergic rhinitis.MethodsAccording to positive sIgE,symptom and sign,we divided the subjects into three groups: AR patients(81 subjects),chronic simple rhinitis patients(21 subjects)and HCs(20 subjects).And then,we subdivided AR patients into two subgroups: specific allergen stimulation subgroup(Artemisia pollen+35 subjects,dust mite+24 subjects)and non-specific allergen stimulation subgroup(other allergen+ 22 subjects).Peripheral blood samples were collected.First,we proposed a simple approach using the eosinophil's intrinsic properties,combined with detection of CCR3 expression,to reliably identify eosinophils.Second,in resting state,we detected expressions of CD63 and Fc?RI in blood eosinophils of AR patients,chronic simple rhinitis patients and HCs by Flow cytometry(FCM).Third,after incubating with Artemisia pollen and dust mite,expressions of CD63 and Fc?RI in blood eosinophils of specific allergen stimulation subgroup,non-specific allergen stimulation subgroup and chronic simple rhinitis patients group were detected by FCM.Taking sIgE as the diagnostic criteria,we evaluated the value of CD63 and Fc?RI in diagnosis of allergic rhinitis by receiver operating characteristic curve(ROC curve)and Spearman correlation analysis.Results1.Absolute numbers of CCR3+ eosinophils of AR group were higher than those of chronic simple rhinitis patients group and HCs group(P<0.05).2.In resting state,the expressions of CD63 in eosinophils in the blood of AR group were enhanced with those of chronic simple rhinitis patients group and HCs group(P<0.05);The expressions of Fc?RI in eosinophils in the blood of AR and chronic simple rhinitis patients were enhanced with those of HCs(P<0.05).3.Specific stimulation by Artemisia pollen and dust mite can upregulate the expressions of CD63 of specific allergen stimulation subgroup(P<0.05);but stimulation by Artemisia pollen and dust mite can't enhance the expressions of CD63 of non-specific allergen stimulation subgroup and chronic simple rhinitis group(P>0.05).We failed to see significant changes in the expressions of Fc?RI in eosinophils of specific allergen stimulation subgroup,non-specific allergen stimulation subgroup and chronic simple rhinitis group by allergen stimulation(P>0.05).4.Areas under the ROC curves of CD63 were from 0.8 to 0.9,but areas under the ROC curves of Fc?RI were from 0.5 to 0.6.After stimulation by different kind of allergens,induction of expressions of CD63 in eosinophils and the level of sIgE are positive related.Conclusions1.Numbers of CCR3+ eosinophils of AR group were higher.2.In resting state,the expressions of CD63 in eosinophils in the blood of AR group were enhanced with those of chronic simple rhinitis group and HCs.The expressions of Fc?RI in eosinophils in the blood of AR group and chronic simple rhinitis group were enhanced with those of HCs group.3.Specific stimulation by allergen can induce the upregulation of CD63 in eosinophils,but Fc?RI did not change so much.4.In eosinophil activation test,using CD63 to diagnose allergen was more valuable,induction of expression of CD63 in eosinophils and the level of sIgE are positive related.
